OBJECTIVES:To detect the physical characteristics of allogenic arteries cross-linked by Dye-mediated photooxidation (DMP) and Genipin (GP), as well as its biological propreties after being treated with allogenic vessel transplant and internal bloodstream, based on the former researches that biological tissue which have gone through different cross-linked methods will show up different biological propreties. Besides, an initial study is done toward the middle and long-term effect of allogenic arteries cross-linked by DMP.METHODS:The thoracic arteries of New Zealand rabbits (with the vessel inside diameter of 4mm), according to different cross-linked methods, were divided into DMP group, GP group, Glutaraldehyde(GA) group, and a control group is also established (which has not gone through any cross-linked treatment). After the arteries were cross-linked with different methods, tissue water content, heat shrinking temperature, breaking tenacity, and biocompatibility which have gone through the external hemolytic test were detected. Each cross-linking artery was transplanted to a new rabbit, the comparative group was orthotopic transplanted(OT). Postoperative animals were feeded for 1 month, the transplanted arteries were taken out to evaluate the biological stability by a semiquantitative counting under microscope. Also to evaluate the cytotoxicity by semiquantitative counting the coverage of endothelial cell(ES) inside the arteries under scanning-electron microscope(SEM). Anti-calcification property was evaluated by measuring calcium content in transplanted arteries through Atomic absorption spectrometry(AAS).Another rabbits which were transplanted arteries cross-linked by DMP, were feeded for 4-6 months, then the transplanted arteries were taken out and repeated the above studies.RESULTS:(1) Tissue water content of cross-linked arteries:control group>DMP group>GP group>GA group. There is no statistical difference between control group and DMP group, between GP group and GA group. In DMP group, the tissue water content of cross-linked arteries was obviously higher than that in GP group(P<0.05).(2) Heat shrinking temperature of cross-linked arteries:GA group>GP group>DMP group>control group. There is no statistical difference between GA group and GP group, between DMP group and control group. In GP group, the heat shrinking temperature of cross-linked arteries was obviously higher than that in DMP group(P<0.05).(3) Breaking tenacity of cross-linked arteries:GA group>GP group>DMP group>control group. There is no statistical difference between GA group and GP group, between DMP group and control group. In GP group, the heat shrinking temperature of cross-linked arteries was obviously higher than that in DMP group(P<0.05).(4) Indices of external hemolytic of cross-linked arteries:GA group>GP group>DMP group. All of them were lower than the standard of American Society of Testing Materials(ASTM) and national(5%). There is no statistical difference between GP group and DMP group. On the contrary may, the statistical differences between GA group and GP group, between GA group and DMP group are apparently(P<0.01). (5) Biological stability of transplanted arteries:control group>DMP group>GP group>GA group. There is no statistical difference among control group, DMP group and GP group. In DMP group and GP group, the biological stability of transplanted arteries was obviously better than that in GA group(P<0.01).(6) The coverage of ES of transplanted arteries:DMP group>GP group>GA group. There is no statistical difference between DMP group and GP group. In DMP group and GP group, the coverage of ES of transplanted arteries was obviously higher than that in GA group(P<0.01).(7) Calcium content in transplanted arteries:GA group>GP group>DMP group>control group. There is no statistical difference between GA group and GP group, between DMP group and control group. In GP group, the Calcium content in transplanted arteries was obviously higher than that in DMP group(P<0.01).(8) Another rabbits which were transplanted arteries cross-linked by DMP, were feeded for 4-6 months, then the transplanted arteries were taken out. Compare with the DP group, it also has the better biological stability, anti-calcification property, and lower cytotoxicity.CONCLUSIONS:(1) The thoracic arteries of New Zealand rabbits (with the vessel inside diameter of 4mm), after gone through different cross-linked treatments, there are such changes as a decrease in tissue water content, increase in heat shrinking temperature and breaking tenacity. Compared with DMP, GP have shown a better physical characteristics in cross-linking the arteries. So it is more convenient for molding and surgical operation.(2) All of indices of external hemolytic of arteries cross-linked by DMP, GP and GA were lower than the standard of ASTM and national(5%). It is suitable for bodies transplanted.(3) Compare with the GP, DMP have shown a better anti-calcification property, but no advantage in biological stability and cytotoxicity. And the anti-calcification property is no statistical difference between GP and GA.(4) The arteries cross-linked by DMP, after transplanted for 4-6 months, also has the better biological properties. In vascular graft field, DMP show a comprehensive application foreground, it is suitable to transplant for a long time.
