ObjectiveThe glioma is the most common malignant tumor in the central nervous system, and the incidence is significantly higher than other intracerebral tumours, and surgery isnot easy to cut, and easy to recurrence and with poor prognosis. Research shows that the epidermal growth factor receptor (EGFR) autocrine secretion is one of the important factors of the gliomas cell growth and infestations, and as its substrate membrane even protein A2 (ANXA 2) play a key role in DNA synthesis and cell proliferation。The purpose of this experiment is the study of the expression of ANXA2 and EGFR in different level of glioma, and the relationship of glioma in different pathological level, to further clarify the gliomas cell division hyperplasia and aggressive molecular mechanism, provides the theory basis for its diagnosis.MethodsThe paraffin-embedded specimens of gliomas were taken from department of neurosurgery in First Affiliated Hospital of Zhengzhou University from May 2008 to Oct 2010, including 36 males,40 females, mean age was 6.5 ages to 83 ages, the averae age is 44.09±2.06 years old. Pathological grading was classified according to the standard pathological grades for glioma.GradeⅠ10 cases, gradeⅡ28 cases, gradeⅢ22 cases, gradeⅣ16 cases. We combined gradeⅠand gradeⅡof glioma to low grade glioma(LGG), and gradeⅢand grade IV to high grade glioma(HGG).The ANXA 2 and EGFR proteins were determined in the normal, the LGG and the HGG by imunohistochemical Streptavidin- peroxidase (S-P)technique. We respectively useχ2 test and Fisher exact probability method to calculate the difference between the normal brain tissue and gliomas, then use Spearman rank correlation analysis to analyze the relevance of ANXA2 and EGFR.Results1. EGFR:As the rise of the pathological level of glioma, the positive expression percentage of EGFR respectively isⅠ30.00% (3/10),Ⅱ42.86% (12/28)Ⅲ68.18%(15/22),Ⅳ87.50%(14/16).The positive of low-grade malignant of glioma is 39.47%(15/38), and the highly malignant is 76.31%(29/38). We can kown that there is significant difference between the expression in statistics.2. ANXA 2:As the rise of the pathological level of glioma, the positive expression percentage of ANXA 2 respectively isⅠ10.00%(2/10),Ⅱ46.43%(14/28),Ⅲ72.72%(16/22),Ⅳ87.50%(14/16).The positive of low-grade malignant of glioma is 42.10%(16/38), and the highly malignant is 78.95%(30/38). We can kown that there is significant difference between the expression in statistics.3. Both the ANXA 2 and the EGFR of glioma organization in protein expression have the difference in statistics, and have closely related with each other.Conclusions1. EGFR do not express in the normal brain tissue.But as the rise of the malignant degree of the glioma, the EGFR's expression level is closely related with it.2. ANXA 2 do not express in the normal brain tissue.But as the rise of the malignant degree of the glioma, the expression level of ANXA 2 is closely related with it.3. There is significant positive correlation between the ANXA 2 and EGFR in the expression of human brain glioma.What's more, they also play a very important role in the occurrence and development of the glioma cells.So EGRF and ANXA 2 may become the effective reference index of diagnosis of the human glioma, and may prove to be the new direction of target-based research and treatment.
