With the development of molecular biology, the incidence of tumor development is a complex multi-step continuous process was recognized. EGFR is a membrane protein tyrosine kinase receptor, through several signal transduction pathways regulating cell function.EGFR plays an important role in glioma cell proliferation. Many clinical studies show that signal transduction pathway of EGFR closely related cancer clinical stage and metastasis. Matrix metalloproteinase -9 (MMP-9) is the degradation of extracellular matrix (ECM) of the major proteolytic enzymes, ECM degradation can restrict tumor cell invasion to the barrier of the surrounding tissue damage. MMP-9 in glioma invasion plays an important role. Ki67 is a marker genes of nuclear proliferation. their expression can reflect the proliferation of tumors, and related to the tumor grade and prognosis. Ki67 is used to judge a reliable method of tumor cell proliferation commonly. Thus,to investigate the relation of the EGFR expression of glioma and proliferation and invasiveness with different levels of EGFR, MMP-9, Ki67 expression of the different level of glioma. Methods: From January.2005 to September.2009, we collected 102 samples of human gliomas and 8 samples of normal brain tissues from the patients, received the operation in the Affiliated hospital of hebei university. The study was focused on the expression of EGFR, MMP-9 and Ki67. Expression of the EGFR, MMP-9 and Ki67 was investigated by immunohistochemistry in 102 specimens of human gliomas. Results: The results showed that EGFR and MMP-9 in 102 patients with glioma were positive 60.78%, 64.71%, while no expression in normal brain tissue. There were significant differences between glioma and normal brain tissue groups (P <0.01); Glioma tissues at all levels of Ki67 proliferation index was significantly (F = 1.00, P <0.01). In addition, EGFR and MMP-9 protein expression was positively correlated (r = 0.75, P <0.01).EGFR expression and the expression of Ki67 was positively correlated (r = 0.983, P <0.05) Conclusion :1 EGFR, MMP-9, Ki67 only expression in glioma, while no expression in normal brain tissue. EGFR, MMP-9, Ki67 express in everyl levels of grade。2 EGFR and MMP-9 expression was positively correlated with EGFR expression Show EGFR expression revealed the invasion ability of gliomas.3 EGFR expression and Ki67 expression was positively correlated, show EGFR and its signal transduction pathway involved in the abnormal proliferation of glioma. | | Keywords/Search Tags: | glioma, EGFR, invasiveness, proliferation, Immunohistochemistry | PDF Full Text Request | Related items |
