Objectives The survey focuses on establishing severe acute pancreatitis animal model. Through dynamic testing the content of plasma endotoxin D-lactic acid,and observating the change of the pancreas, ileum pathological form, the author discusses the role of endotoxin and D - lactic acid in the early diagnosis of severe acute pancreatitis with intestinal injury.Methods 70 specific pathogen free (SPF) wistar male rats were divided into three groups randomly: control group (Control),Sham-operated group(Sham),and pancreatitis group.Sham and pancreatitis group are divided into 6 hours, 12 hours and 24 hours three time points.Pancreatitis was induced by injection of 5% Na-Tc under membrane of pancreas. The rats were killed in sixth hours'time after being operated in Sham-operated group. The rats were killed in the sixth, twelfth and twenty-fourth hour after Na-Tc injection in Six hours after pancreatitis group, Twelve hours after pancreatitis group and Twenty-four hours after pancreatitis group respectively. The content of amylase in serum was measured by holo-automatic biochemical analysator. Pancreatic tissues were observed by hematoxylin-eosin (HE) staining and counted for the pancreatic tissues and histological scores. At same time detected the concentration of Endotoxin ( ET) in plasma with ELISA method. Adopting enzymology spectrophotometry detection of serum D - lactic acid content .All the data were analyzed by statistics software SPSS17.0. Normal distribution measurement data shown as means±standard deviation ( x±s). One-way ANOVA method was applied to comparison among groups. And using S-N-K method for binary comparison. Pearson correlation analysis was applied to measure the relevant relations. The inspection standards was alpha = 0.05.Results We successfully established severe acute pancreatitis animal model. The rats of pancreatitis group exhibited a high amylase and severe pancreatic pathological damage after modeling.The ileum appears pathological damage after building mode 24h. There are non- significant difference (P>0.05) in sham group and control group and sham group each time in Pancreas pathological scorein;but in different periods,SAP groups was significantly different compared with sham-group and blank group;SAP group has significant differences compared with each other.The content of endotoxin and D-lactic acid in sham-operation group each time and blank group has no significant differenc(eP>0.05);but in different periods,SAP groups was significantly different compared with sham- group and blank group(P<0.05);SAP group has significant differences(P<0.05) compared with each other. The contend of SAP group of plasma endotoxin, D-lactic acid and pancreas pathological score exists linear correlation. This kind of correlation also exists between the content of plasma endotoxin and D-lactic acid.Conclusions We successfully established the severe acute pancreatitis models by using the method of injecting 5% Na-Tc under membrane of pancreas. The SAP intestinal damage of plasma endotoxin and D - lactic acid are increased in the early period, and gradually increased with the increase of pancreas pathological injury. Therefore,detection plasma endotoxin and D-lactic acid can be used as the current theoretical basis of early diagnosis in severe acute pancreatitis with bowel function injury.
