Polysaccharides is not only the energy resource or fabric materials, but also it has the informational function that can compare with nucleic acid and protein, it also has the biological function that participate in every life activity, antivirus, ant infection, resist radiation and so on. The angustifolia has large grow area in sinkiang, so extracted Polysaccharides from oleaster has extensive exploder foreground based on economy and decrease the resource losing. In this paper, the angustifolia is produced in sinkiang.The main research object is to research several methods of extracting angustifolia polysaccharide. Using the traditional hot water extraction, microwave-assisted extraction method, ultrasound-assisted extraction method, respectively orthogonal test, is to fing the optimum extraction. The extracting of polysaccharides from angustifolia was precipitated by ethanol. Polysaccharide is purified by n-butanol, 20%trichloroacetic, n-butanol-20% trichloroacetic method to remove protein, and then followed by DEAE-52 column chromatography. Polysaccharide is shown to be a single homogeneous component by HPLC and SephadexG-100 column chromatography. the ant oxidation activities of polysaccharides is studied. The results showed that:1.Traditional hot water extraction of polysaccharide optimum conditions are as follows:extraction temperature 80℃, extraction time 1 h, water:angustifolia=35:1, Extraction times 2.The extraction rate is 5.1%.Microwave-assisted extraction of Angustifolia polysaccharide optimum conditions are as follows:Extracting power 450 W, extraction time 2.5 min, water:angustifolia=25:1, The extraction rate is 8.98%.Ultrasonic extraction auxiliary angustifolia polysaccharide optimum conditions are as follows:Ultrasonic power 120 W, extraction time 40 min, extraction temperature 40℃, water: angustifolia=40:1, The extraction rate is 7.5%.Considering various factors, microwave-assisted extraction is the optimal extraction method.2. The extracting of polysaccharides from angustifolia were precipitated by ethanol. Polysaccharide is purified by n-butanol,20%trichloroacetic, n-butanol-20% trichloroacetic method to remove protein, and compared with this three methods. The results showed that:it is the best way to remove protein from Polysaccharide using n-butanol-20% trichloroacetic method, the polysaccharides losing rate is 35.23% and the protein removed rate is 13.23%.3. The polysaccharides that has been removed protein followed by DEAE-52 column chromatography, and we obtained two kinds of polysaccharides.
