Objective:To explore the Rock2 regulatory mechanism on cell cycle protein Cdc25A and to study the growth effect in hepatoma carcinoma cells.Methods:After transfection of Rock2-siRNA into hepatoma carcinoma Huh-7 and HepG2 cells, flow cytometer and MTT assay were used to observe the changes of cell cycle and cell growth. In response to UV induced DNA damage, Cdc25A protein expression was detected by Western blot.Then the changes of Rock2 and Cdc25A were tested by Western blot, after the transfection of Rock2-siRNA. Finally, Cdk2 activity was detected in Rock2 knockdown cells and the normal cells. The results were analysed to recognize the Rock2 regulatory mechanism on Cdc25A in hepatoma carcinoma cells.Results:Rock2 serves the protective effect on cell survival. The expression of Cdc25A reduced following the decline of Rock2 after Rock2 interference. When exposed to UV damage, the reduction of Cdk2/Cyclin E activy was accompany with the decrease of Cdc25A.Conclusion:Rock2 plays a regulatory role on cell cycle checkpoint Cdc25A in hepatoma carcinoma cells.
