Objective:This experiment with big rat hippocampal infarction, give rehabilitation training for model of rats were observed after learning and memory capacity, the hippocampus histology and SDF-1 alpha positive cells expression changes.Methods:54 healthy SD rats (male, weigh 200-250g) adopting digital random method to divide into three groups:JiaShouShu group, the model group and rehabilitation group. Three groups were divided into training 3 days, training 7 days, training 14 days three time points. Each time point for only 6. Using photochemical legal system model of the hippocampus infarction prepared. After the success of Morris water to model a maze training, daily training three times, each time middle-late early. The end of therapy at different time points after testing the following index:(1) avoid dark application method to detect learning and memory capacity; (2) HE dyed observation the hippocampus-histological change; (3) using immunohistochemical method to detect the hippocampus SDF-1 alpha positive cells expression.Results:(1) each rat camera obscura test results:avoid JiaShouShu group than with model rats, learning and memory, and ability to drop, a statistically significant difference (P<0.01); Compared with model group, rehabilitation group study memory capacity of restore somewhat, and each time point group compared with model, a statistically significant difference (P<0.05). (2) HE dyed display:JiaShouShu group of neurons in the hippocampus aligned, did not see the pathological changes. Model group visible structure change, the decrease in the number of cells, cells are arranged disorder, nuclei become pyknotic, cytoplasmic hyperchromatic. Recovery group compared with model group, the hippocampus structure change relatively light. (3) group the hippocampus SDF-1 alpha positive cells expression results show that:JiaShouShu group in SDF-1 a brain cells express positive alpha. And JiaShouShu group 3 days,7 days,14 days comparison, model group ischemia area SDF 1 alpha positive cells increases, the expression was statistically significant difference (P<0.05). And the model group 3 days,7 days, 14 days of comparison, rehabilitation SDF-1 alpha positive cells more apparent, express increase was statistically significant difference (P< 0.05).Conclusion:(1) rehabilitation training can promote learning and memory capacity recovery; (2) big rat hippocampal infarction light-microscopy neuron structure in the hippocampus in the marked changes, rehabilitation training can reduce the damage brain tissue. (3) rehabilitation training can promote large cerebral ischemia SDF-1 alpha area, which could express positive cells is rehabilitation training to promote cerebral ischemia in rats mechanism of learning and memory, one of the ability to increase.
