Spleen yang deficiency diarrhea is one common type of diarrhea, According to CTM theory,the fundamental reason for diarrhea is most often the insufficiency of spleen.Chronic diarrhea can lead to spleen Yang deficiency,even can cause deficency of the kedney.Professor Liu ShenLin of Jiangsu hospital of Chinese medicine adopt " WenPiShiChang method" to treat Spleen yang deficiency diarrhea based on years of clinical experience. Plenty of clinical cases prove that wenpishichang formula can work. In order to study the effect of WenPiShiChang formula on spleen Yang deficiency diarrhea and its mechanism,we made the following experiment.Part1 The influence of WenPiShiChang formula on gastrointestinal function.At the first section,100% rhubarb decoction was given intragastrically and reserpine was given by subcutaneous injection,in order to build a model of mice spleen-yang deficiency. After building the model,we give wenpishichang formula by i.g to observe the influence of WenPiShiChang formula on gastrointestinal function.The results suggest that WenPiShiChang formula could increase the rate of gastric emptying in mice,reduce rate of carbon powder going in mice intestine, enhance the inhibitory effect of dopamine on gastrointestinal function and inhibit neostigmine inhibition of gastrointestinal function.Part2 The anti-inflammation and Analgesic effect of WenPiShiChang formulaIn this part,we study the anti-inflammation and Analgesic effect of WenPiShiChang formula.At the first section,60 ICR female mice were randomly assigned,then, WenPiShiChang formula were given intragastrically.The analgesic effect of WenPiShiChang formula was studied by mouse acetic acid-induced twisting experiment and hot plate test. Models of acute inflammatory reaction of xylene-induced mouse ear edema and capillary permeability induced by acetic acid were performed to study the antiinflammatory.The results prove that ear edema resulted from xylene and capillary permeability induced by acetic acid were significantly inhibited by WenPiShiChang formula,and WenPiShiChang formula could produce analgesic effect.Part3 The effect of WenPiShiChang formula on digestive enzymes of spleen-yang deficiency diarrhea rat modelsAt this section, SD rats were taken.The spieen-yang deficiency diarrhea model of rats were induced by 100% rhubarb decoction.Rats with spleen-yang deficiency diarrhea were randomly assigned. Then, WenPiShiChang formula were given intragastrically.After the last treatment,we taked blood from orbital vein plexus. The AMS,LDH and SDH in serum were determined at last. The result display that WenPiShiChang formula obviously increased AMS and LDH activity in serum.At the same time WenPiShiChang formula decreased SDH activity in serum.Part4 The influence of WenPiShiChang formula on gastrointestinal hormones of spleen-yang deficiency diarrhea rat modelsAt this section,SD rats were taken.The spleen-yang deficiency diarrhea model of rats were induced by 100% rhubarb decoction.Rats with spleen-yang deficiency diarrhea were randomly assigned. Then, WenPiShiChang formula were given intragastrically.After the last treatment,we extracted gastrointestinal hormones from intestines. The MTL,GAS and SS were determined at last. The result display that the levels of GAS and SS in gastrointestinal tract of spleen-yang deficiency diarrhea rats was lower than the levels of gastrointestinal hormones in normal rats,the level of MTL in gastrointestinal tract of spleen-yang deficiency diarrhea rats was higher than normal rats. WenPiShiChang formula obviously increased GAS and SS in gastrointestinal tract.At the same time WenPiShiChang formula decreased MTL in gastrointestinal tract.Part5 The affect of WenPiShiChang formula on immunologic function and pathology of mouse with Spleen-yang Deficiency DiarrheaIn this part,we observed the effection of wenpishichang formula on T-lymphocytes subgroup (CD4+,CD8+,CD4+/CD8+) in mice with spleen-yang deficiency diarrhea. The method we used to copy the model of spleen-yang deficiency diarrhea,is as it was before. One hour after the last treatment,eye blood samples were collected in each groups.The T-lymphocytes subgroup were detected by FCM.At the same time, we taked gastrointestinal tract from mouse,then the gastrointestinal tract were stored in formalin immediately.According to routine method, make the paraffin, at last the variations of the pathology were observed under electrical microscope.But,the Structure of intestinal mucosa did not have significant changes.On even ground,we observed the effection of WenPiShiChang formula on sIgA in serum of rats with spleen-yang deficiency diarrhea.The results show that The levels of CD4+and CD4+/CD8+in mice with spleen-yang deficiency diarrhea were lower than the normol control group.The large doses of wenpishichang formula can raise the level of CD4+and CD4+/CD8+.The content of sIgA in serum of rats with spleen-yang deficiency diarrhea was lower than the level of sIgA in the normal rats. WenPiShiChang formula can improve the level of sIgA.Conclusion:WenPiShiChang formula can inhibit accentrated movement of gastrointestinal when spleen-yang deficiency diarrhea occurs.At the same time, WenPiShiChang formula has significant anti-inflammatory and analgesic effects.The mechanism of WenPiShiChang formula to inhibit spleen-yang deficiency diarrhea maybe related to the following factors:First, Wenpishichang formula can correct the parasecretion of digestive enzymes;Second,it can regulate the secretions of gastrointestinal hormones;Thirdly, Wenpishichang formula have the action of increasing immune function.
