Background and purposeEsophageal cancer is one of the most common malignant neoplasms in the digestive system.It was the fourth in the death rate of malignant tumor in our country. The occurrence and development of tumor is a process of chages involving multi-stage, multi-step, multi-factor and polygene.Pim-3 is a member of a conserved family of kinases originally identified as proto-oncogenes, and expression of these kinases is stimulated by numerous growth factors and cytokines. Pim-3 expression is enhanced in several cancer tissues, particularly those of endoderm-derived organs, including liver, pancreas, colon, and stomach. Minichromosome maintenance protein 7 (MCM7) is involved in replicative licensing and synthesis of DNA. MCM7 expression levels have been examined in several human cancers, including prostate cancer, colorectal cancer, neuroblastoma, cervical and hypopharyngeal carcinoma。Recent reports propose that regulation of MCM7 protein expression may be a marker of cell proliferation along with Ki-67, which is highly expressed in proliferating cells.The principal objective of this study was to characterize the expression of Pim-3 and MCM7 and to analyze their relationship to clinicopathological parameters in patients with esophageal squamous cell carcinoma(ESCC).Materials and Methods(1)77 esophageal carcinoma tissues were collected in An Yang cancer hospital,70 adjacent atypical hyperplasia tissues and 46 normal esophageal tissues were collected in the first hospital affiliated to Zhengzhou University.(2)The expression of Pim-3 and MCM7 were detected by the SP immunohistochemical method in 77 cases of ESCC,70 adjacent atypical hyperplasia and 46 normal esophageal tissues.The correlations between the expressions of Pim-3 and MCM7, and their relationships with the clinicopathologic features of ESCC were analyzed.(3) All of the experimental data was performed with statistical package SPSS 13.0 software.The compassion of positive rates applies Mann-Whitney U Test or Kruskal-Wallis H Test.Association between parameters was assess by Spearman correlation analysis.Statistically significant level was considered as "a=0.05".Result(1)The expressions of Pim-3 in ESCC tissues were significantly higher than those in adjacent atypical hyperplasia and normal esophageal tissues (93.51%,24.29% and 13.04%,H=113.160, P<0.001). The expressions of Pim-3 were positively correlated with the TNM classification (rs=0.386, P<0.05) and lymph node metastasis (rs=0.460, P<0.05)(2)The expressions of MCM7 in ESCC tissues were significantly higher than those in adjacent atypical hyperplasia and normal esophageal tissues (93.51%,62.86% and 52.17%,H=64.507, P<0.001). The expressions of MCM7 were positively correlated with the TNM classification (rs=0.393, P<0.05),lymph node metastasis (rs=0.271, P<0.05). the carcinoma differentiation (rs=0.421,P<0.05) and adventitial invasion (rs=0.317,P<0.05)(3) Spearman correlation analysis found that the expression of Pim-3 in esophageal squamous cancer tissue had no correlation with that of MCM7 (rs=0.116, P=0.316).Concolusion(1)Overexpression of Pim-3 and MCM7 are found in ESCC tissues, and their expressions are positively correlated with the TNM classification and lymph node metastasis, they may play an improtant role in the carcinogenesis and aggression of ESCC.(2) The expressions of MCM7 were also positively correlated with the carcinoma differentiation and adventitial invasion, MCM7 can be used as a marker of cell proliferation of esophageal squamous cell carcinoma.(3)But significance of detection of Pim-3 and MCM7 in ESCC need further exploration.
