Objective:To observe the inhibitory effect on growth and metastasis of BuShenShuGan decoction on mice bearing Lewis lung cancer (LLC) and investigate its underlying mechanism.Methods:Forty C57BL/6 mice that had been successfully established Lewis lung carcinoma model were randomly assigned to four groups:model group,DDP group,Chinese herbal medicine group,combined group. Model group was given intragastric administration with saline 0.4ml everyday; DDP group was given intraperitoneal injection of cisplatin solution 0.5ml (containing DDP 0.1mg) on d1,d3,d5; Chinese herbal medicine group was given intragastric administration with BuShenShuGan decoction 0.4ml/d (containing raw materials 0.7g); combined group was given intragastric administration with chinese medicine and intraperitoneal injection of cisplatin solution which was the same as chinese herbal medicine group and DDP group; all the interventions lasted 21 days. The mental state, diet and activities of mice were observed everyday, body weight and tumor volume measured every two days. All the mice were sacrificed in d22,tumor weight and numbers of spontaneous lung metastasis were measured; the expression of micro vessel density (MVD),vascular endothelia growth factor (VEGF) protein and matrix metalloproteinase-9 protein (MMP-9) were detected by immunohistochemistry.Results:1,Compared with model group, the treatment group could significantly inhibit tumor growth, the difference was significant (p<0.01). Compared with chinese herbal medicine group, the average tumor weight of combined group decreased significantly (p<0.01).2,Compared with model group, the treatment group could significantly inhibit the spontaneous pulmonary metastasis (p<0.05 or p<0.01); Compared with DDP group and Chinese herbal medicine group, the number of spontaneous lung metastasis of combined group was significantly Lower (p<0.05).3-. After the experiment finished, compared with model group, the weight of DDP group decreased (p<0.05); Chinese herbal medicine group and combined group were significantly increased (p<0.05), there was no difference between chinese herbal medicine group and combined group (p>0.05). Compared with model group, tumor volume of every treatment group was significantly smaller (p <0.05); compared with DDP group and chinese herbal medicine group, though the tumor volume of combined group was smaller, but the difference was not significantly(p>0.05).4,Compared with model group, MVD of all treatment groups decreased significantly(p<0.05 or p<0.01); compared with DDP group and chinese herbal medicine group, combined decreased significantly (p<0.05). Compared with model group, VEGF expression of the treatment groups significantly decreased (p<0.05 or p<0.01); compared with DDP group, the expression of combined group was significantly decreased (p<0.05).5,Compared with model group, the expression of MMP-9 of all treatment groups decreased significantly (p<0.01); compared with DDP group and chinese herbal medicine group, combination group decreased significantly (p<0.05).Conclusion:1,BuShenShuGan decoction could significantly inhibit growth and spontaneous lung metastasis of Lewis lung cancer, and there was synergistic effect with cisplatin.2,BuShenShuGan decoction could down-regulate the expression of VEGF and MMP-9, this may be one of the mechanisms of the inhibition of tumor growth and metastasis.3,BuShenShuGan decoction could improve the quality of life, reduce the side effects caused by chemotherapy such as weight loss et al.
