Objective:To explore the expression of interleukin-12 and interleukin-23 in intestinal mucosal of patients with crohn's disease and its relationship with disease severity.Methods:Collected surgical intestinal mucosa specimens from 30 crohn's disease patients, the experiment set CD lesions bowel section group, CD normal bowel section group and normal controls three groups. All CD patients'CDAI scores are in active period,30 cases normal controls are surgical resection of the normal colonic organization of non-inflammatory bowel disease. Using immunohistochemistry detection IL 12 and IL-23 protein expression quantity, comparing expression level difference between three groups, evaluation the relevance between expression of cytokines and the disease clinical seerity(CDAI) and endoscopic intestinal mucosa damage degree (CDEIS).Results:Immunohistochemical analysis results found IL-23pl9, IL-12 protein positive reaction is tan-yellow grain, mainly distributed in cytoplasm, part of cell membrane shading. CD patients IL-23p19, IL-12 positive cells distributed mainly mucosa lamina propria, in the submucosa,muscularis and serous layer also have many positive cells express, epithelial accidentally see expression. Statistical analysis found that CD lesions bowel section group, CD normal bowel period of IL-23p19 positive rate were higher than normal controls bowel section group, a statistically significant difference, CD lesions bowel section group and CD normal bowel period there was difference between groups too. Three groups'IL-12 were no difference. IL-23p19 positive expression and CD of the severity of the disease clinical correlation(CDAI) and endoscopic intestinal mucosa damage degree (CDEIS)has no significant correlation.Conclusion:IL-12 is not elevated in crohn's disease damaged intestinal mucosal;IL-23 is elevated in crohn's disease damaged intestinal mucosal,which means IL-23 plays a role in the process of crohn's disease mucosal immunity,and has a relevant value with the diagnosis of CD.
