Study On Extraction Of Deer Collagen And Therapeutic Effects Of Collagen On Osteoporosis

Purpose: Extract the collagen from the deer bone and antler plate, study on the properties and pharmacological activity of collagen of deer bone and antler plate by enzyme hydrolysis.Methods: The collagen was extracted using hot water from deer bone and antler plate. A monofactor test and the orthogonal experiment were developed to determine the optimum extraction process. We extracted bilateral ovaries to establish osteoporosis model, treated after 7 days and for consecutively 90 days, then observed the effects of collagen of deer bone and collagen of antler plate on bone mineral density, serum marker, bone histomorphometry, bone mechanics, body weight and organ index.Result: 1,The optimum preparation technology of deer bone was obtained as follow: material-liquid rate was 1:5(g/ml), boiled 3.5h, extracted 3 times; Adopted SDS-PAGE to ensure the trypsin-substrate rate was 1:5000 and hydrolyze 1 hour, collagen yield was5.11%, protein content was80.59%.2,The optimum preparation technology of deer bone was obtained as follow: material-liquid rate was 1:5(g/ml), boiled 5h, extracted 4 times; Adopted SDS-PAGE to fix the trypsin-substrate rate was 1:10000 and hydrolyze 1.5 hour, collagen yield was22.31%, protein content was66.32%.3,Compared with osteoporosis model group, after treating by collagen of deer bone and collagen of antler plate, bone mineral density, ALP and Hyp in serum, bone histomorphometry, and bone mechanics are significant differences, level of serum E2, body weight and organ index have no statistical significance.Conclusion: 1,The extraction ways of collagen of deer bone and antler plate is simple, extraction rat is higher.2,Collagen of deer bone and antler plate can treat osteoporosis rats.
