Micro-Channel Electrophoresis chip (MCE chip) is a novel micro-analysis tool based on Capillary Electrophoresis technologies. The samples are electrophoresed in the micro-well and micro-channel networks on the substrate of glass, fused silica, and polymers, etc. fabricated by the Microelectronics technology, and are detected by Optical, Electrochemical Methods, etc. MCE chip is a interdisciplinary technology that roots from Analytical Chemistry, Biochemistry and the technologies of Microelectronics, which provides a new platform :br separation and analysis in the fields of Life Science, Medicine, and Materia Medic a, and has been used for more and more applications.The development and principle of MCE chip were reviewed, the development of MCE chip detection system in our laboratory was iiscussed in this dissertation. The effect of the separation medium for DNA analysis was tested on the chip detection system, and the Detection of Isoniazid Resistance-associated Mutations in M. tuberculosis was studied on this system, some good results were obtained.The topics of this dissertation are presented below:1. The proposal and development of Capillary Electrophoresis and MCE chip were reviewed.2. The fundamental principle of MCE baj ed on Capillary Electrophoresis and the development of MCE chip del ection system were described.3. The principle of DNA separation with polymer solution by CE and MCE was described. The research on separation medium of polymer was reviewed. The separation effect of the different concentration solutions of two kind of polymers with different Molecular Weight was studied on the detection system, a separation medium with small amount of montmorrillonite clay was also studied.4. The detection of Isoniazid resistarce-associated mutations in M. tuberculosis was performed on the chip detection system, and some goodresults were obtained.It is displayed in this dissertation that the realization of DNA sequencing on the MCE chip need further works yet, however, the detection of resistance-associated mutations on this system is fast and sensitive, and could be potentially applied for clinical diagnosis.
