| Objective:To observe the effect of hyperuricamia on insulin sensitivity of rat.Methods:To establish hyperuricemia model of rat with yeast and adenine, and conduct hyperinsulinemic-euglycemic clamp test in No.4,6,8,10,12 week and calculate glucose infusion rate (GIR) to evaluate insulin sensitivity of rats, measuring serum uric acid (UA), fasting blood glucose (FBG), fasting insulin (FINS) with angular vein blood every week. In No.6,8,10 week to lift the model groups of rats with high uric acid status, and observe the target again.Results:Successfully established hyperuricemia model of rat, the GIRof experimental group appears decreased in the 4th week, then continue at a low level, compared with the control group, the differenciate were statistically significant (P<0.01); with the decline of insulin sensitivity of rat, the fasting blood glucosep of sustained-hyperuricemia group and high-purine fed-for-10 weeks group appear increased, compared with the control group significantly (P<0.01);) The insulin appeared decreased (P<0.01) in the 6th week compared with the control group, then continued to decrease. High-purine fed 8-week group were at normal blood glucose levels, high-purine fed 8-week group appear hyperglycemia in the 11th week,the two groups appear lower insulin levels in the 6th week,the insulin levels become a recovery trend after lifted high uric acid, but failed to reach normal levels.Conclusions:Sustained hyperuricemia can reduce the insulin sensitivity of rats and cause hyperglycemia and insulin levels decreased, lifting of the state of hyperuricemia earlier can prevent diabetes. |
