| Zhimu is the dry rhizome of Anemarrhena asphodeloides Bge. which belongs to Liliaceae. It is an important clinical medicinal material embodied in Chinese Pharmacopoeia. Genetic factors, environmental factors, cultivation practices, and many other factors are affecting the quality of Zhimu. In the legioned factors that influence the quality of cultivate Zhimu, we selected the variation of active ingredient content of different drying processing methods and different aged Zhimu as subject, and definituded the changing trend rule. We also compared the effective ingredients in different parts of Zhimu and validated that to remove the root is justified. At the same time, for a general view of the quality of Zhimu in different medical institutions of Beijing, we collected 35 Zhimu samples from hospitals and clinics in Beijing, and determined the active ingredient contents of them to analyse and compare the quality of Zhimu from different medical institutions. All the results are as follow:(1),We dried Zhimu by different methods, including drying in the sun, drying in the shade, drying in the oven, far infrared drying and microwave drying. And we used three temperatures—80℃,60℃,40℃respectively when drying in the oven. Then we compared the ingredient content of Zhimu by different drying mothods wth sarsasapogeni,Anemasaponin AⅢ,neomangiferin,mangiferin in HPLC as target. HPLC-ELSD method of content determination of sarsasapongenin and Anemasaponin AⅢwith internal standard was set up. Precision stability and recurrence are all up to the mustard. HPLC-UV method of content determination of mangiferin and neomangiferin was set up. The calibrate equation of neomangiferin is:y=2841.4x-24.044, r=0.9999 (linearity:0.1047ug~1.2567ug); And that of mangiferin is:y=3745.9x-156.08, r=0.999 (linearity: 0.107ug~0.642ug).The content of sarsasapogenin,Anemasaponin AⅢand mangiferin in Zhimu dried quikly with drying instruments is higher than that dried in sun and in shade. And the tree compositions'variation in Zhimu with different drying methods is similer. The content of tree compositions in Zhimu that dried in the oven by 80℃,dried with far infrared drying method and dried with microwave drying method is the highest, that dried in the oven by 60℃and 40℃is lower, and dried in the sun or in the shade is the lowest. The variation of neomangiferin is almost contrary to that of mangiferin, the content of which is highest in Zhimu that dried in sun and in shade, lower after dried with drying instruments. Considering the active ingredient preservation, the pros and cons of different drying methods in order is: microwave drying> far infrared drying> drying in the oven> drying in the sun≈drying in the shade.Except mangiferin, the active ingredients content in different parts of Zhimu is the highest inside the rhizome, lower in the coat a nd the lowest in roots. The content of mangiferin is the highest in roots, lower inside and the lowest in the coat. It was validated that to remove the roots as non-medicinal parts is justified.From the studies on different aged Zhimu which were separately collected in May and November, we found that the content of sarsasapogen in four-year aged Zhimu was the highest, and it increased gradually with age. But it got down in the perennial ones, and it was approximately as much as that was in three-year aged Zhimu. It was higher in the samples collected in May than that collected in November. In Zhimu that collected in May, The content of Anemasaponin AⅢincreased gradually with age and was the highest in four-year aged Zhimu, approximately as much as that was in the perennial ones. But we didn't find Anemasaponin AⅢin the samples collected in November. The content of mangiferin is the highest in four-year aged samples and that in the other samples were almost the same. The content in May samples was higher than that in November. The content of neomangiferin is the highest in three-year aged samples, lower in four-year aged samples and that in the other samples were almost the same. The content in November samples was higher than that in May. Therefore, Zhimu should be harvested in three or four years.(2),Collecting Zhimu samples from 35 hospitals and clinics locatied in 11 regions of Beijing, with sarsasapogenin,Anemasaponin AⅢ,neomangiferin,mangiferin as target, we compared the quality of Zhimu slices from different medical institutions. We found that the content of active ingredient in Zhimu slices was very different among the medical institutions. But the value of the content of sarsasapogenin,Anemasaponin AⅢ,neomangiferin and mangiferin was not related to the levels of the medical institutions.The distinctness of this article has the following aspects:(1),To reveal the variation trend of Zhimu processed with different original processing methods and harvested in different period by multiple indicator analysis.(2),To investigate and research Zhimu slices from hospitals and clinics of Beijing and generalize the quality of Zhimu slices from different medical institutions. |
