Background and pourpose:To investigate the expression of matrix metalloproteinase-2,9 (MMP-2, MMP-9) and tissue inhibitor of metalloproteinase 3 (TIMP-3) in transitional cell carcinoma of bladder about the expression intensity and distribution. Discussion with TCCB of pathological grade and clinical stage, elevated MMP-2, MMP-9 and TIMP-3 expression in cancer tissue strength and tissue distribution changes. Through this experiment TCCB of MMP-2, MMP-9, TIMP-3 were determined, presumably in the development of bladder transitional cell carcinoma, metastasis in the process of role, in order to conduct the clinical prognosis of TCCB of to determine and carry out the treatment of TCCB provides a new way.Methods:Using a method of immunohistochemical stains and a high-resolution color graphic analysis of medical systems,the expressions of MMP-2,MMP-9 and TIMP-3 were analyzed in human samples from 38 cases of bladder carcinoma(tumor group),10 cases of normal bladder(control group).Result:1.MMP-2, MMP-9, TIMP-3 was significantly higher in TCCB than in adjacent normal bladder mucosa (P<0.01). normal bladder epithelial MMP-2, MMP-9 were negative expression, TIMP-3 expression is low.2.MMP-2, MMP-9 expression in tumor epithelial cells and expression of non-uniform intensity distributied, with mesenchymal tissue, blood vessels near the surface and deep tissues of bladder tumor invasion into the leading edge of positive cells express a strong intensity. TIMP-3 expression in tumor cells and expression of relatively intensity distribution is uniform. Bladder normal tissues and bladder cancer tissue MMP-2, MMP-9 and TIMP-3 optical density value of the difference was significant.3.MMP-2 and MMP-9 were highly expressed in tumor samples and their expression rates were increased with grades and stages. TIMP-3 was also highly expressed in tumor samples and its expression rates was increased with grades too but not with stages.Conclusion:1.MMP-2 and MMP-9 were highly expressed in tumor samples and their expression rates were increased with grades and stages. TIMP-3 was also highly expressed in tumor samples and its expression rates was increased with grades too but not with stages.2. These results suggest that MMP-2, MMP-9 and TIMP-3 play an important role in the invasion step of transitional cell carcinoma of bladder. Detection the expressions of them may become a new mean for the diagnosis of urothelial carcinomas. 3. MNIP-2,MMP-9 and TIMP-3 are better molecular markers in the assessment of bladder transitional cell carcinoma invasion and metastasis, and they can be used to predict the prognosis of tumor. They Also offer a potential strategies to treatment of bladder transitional cell carcinoma.
