| Background and Purpose With changes in lifestyle and diet, the morbidity of increase in the incidence of diabetes is increase, diabetic retinopathy in type 2diabetes also will increase. Diabetic retinopathy is one of four general blindness disease, is one of the five major causes of death, serious effect on patients quality of life and socio-economic development. The pathogenesis of diabetic retinopathy is very complicated, its precise mechanism is not clear yet, but the individual susceptibility of diabetic retinopathy more and more attention, It is a hot topic for the development of diabetic retinopathy study on gene aspect in foreign and Chinese research. This study aims to analyze HLA-DR expression in the type 2 diabetes mellitus patients with proliferative diabetic retinopathy and effect to diabetic retinopathy on the occurrence and development, and to explore the genetic risk factor for PDR, provide scientific basis of prevention and the clinical treatment, as well as provide the basis for overcoming the social and psychological problems to a large number of DM patients。Method Randomly selected in the First Hospital of Shanxi Medical University clinic and hospital type 2 diabetes mellitus in treatment of duration of Shanxi Province and they course of disease are more than 10 years without complication of diabetic retinopathy 30 cases as the patients group, select patients with T2DM complicated by PDR 30 cases as control group,30 cases healthy people as control group for the study.4ml blood collected and the extraction of DNA, on the HLA-DR3, DR7, DR11 gene PCR amplification and determination of gene frequency. Diagnosis confirmed by clinical examination, including fundus examination and OCT examination and FFA, etc. Check. On the use of SPSS 16.0 package for statistical analysis to the statistic,Ï< 0.05 as statistically significant. Results HLA-DR7 expression in T2DM patients without the DR and course of disease more than 10 years yor an average of88.53±8.31, and in T2DM patients with PDR average 18.88±0.82; HLA-DR3, DR11 the average frequency in the PDR were 80.55±3.55,84.15±6.51, the average frequency of the normal control group were 14.48±0.182,27.86±1.415. HLA-DR3, DR7, DR11 in NDM group, DM group, and PDR comparison of expression of each group were p<0.001, the difference was statistically significant. Conclusions HLA-DR7 in Shanxi Han T2DM duration> 10 years with no complication of PDR patients are increased expression. HLA-DR7 the occurrence and development of the DR may have protective effects; HLA-DR3 expression in the PDR increased significantly prompted PDR Genetic susceptibility tendency of gene susceptibility HLA-DR3. |
