| Objective:To observe the changes of the tumour necrosis factor alpha (TNF-α),IL-1,PGE2 in the plasma in the synovial of the collagen II induced arthritis(CIA) in rats,and explore the role of TNF-α,IL-1 and PGE2 in rheumatoid arthritis;To observe the influence of the Sanhuwan on TNF-α,IL-1,PGE2 concentrations in the plasma,and explore the mechanism of the Sanhuwan in the treatment of rheumatoid arthritis disease.Methods:Sprague-dawlay rats(n=32)were randomly divided into four groups:①Normal control group (Normal group, n=8);②Model control group(Model group,n=8);③Yishenjuanbiwan treated group (YSJBW group, n=8);④Sanhuwan treated group (SHW group, n=8).The rat experiment arthritis model was established by left foot subcutaneous injection of II collogen and complete Freund's adjuvant to the Model group, the Yishenjuanbiwan group and the Sanhuwan group, but the Normal group was not injected. Rats of the Sanhuwan group and the Yishenjuanbiwan group were respectively treated twice everyday with Sanhuwan and Yishenjubiwan after 7 days when they were immunized and the Sanhuwan and Yishenjuanbiwan groups were treated twice everyday, and the model group were given distilled water,and the normal group drank freely.after 14 days,The blood of the normal group and that of the model group,the Sanhuwan group and the Yishenjuanbiwan group was gotten by breaking their heads in the 21th,The plasma TNF-α,IL-1 and PGE2 content of the four groups were tested with the radioimmunologic assay,The symptom of arthritis was evaluated by the method of arthritis index integra.Results:(1)General condition observtions:The diet of the model group rats reduced,and the mean weight of the model group was lighter than that of the Sanhuwan group and the Yishenjuanbiwan group.There were some symptoms in the model group,for example,dry and dall hair,but there were not these sympoms in the normal group,the Sanhuwan group and the Yishenjuanbiwan group.(2)24 rats occurenced arthritis,and the incidence of arthritis was approximately 100%.in the second day when the rats werw immuned with II collogen,we can saw the original sweeling of the left feet of all rats achieving high tide,in 9th day,the right feet of a part of rats occurenced subsequencent swelling after 14th day,the swelling were steading.(3)Result of the evaluating integra of arthritis index:The integra of arthritis index of the model group,Sanhuwan group and Yishenjuanbiwan group was increasing with the time elongation,but there was no significantly diffirence on integral of arthritis index among by comparison 14 days ago The integral of arthritis index.arthritis of the model group was higher than that of the Sanhuwan group and Yishenjuanbiwan group 17 days later(p>0.01);the integra of arthritis index of the model group drooped slowly.(4)Result of the radioimmunologic assay:After the model was estalished for 21 days,the plasma TNF-α,IL-1,PGE2 contents of the model group,the Sanhuwan group and the yishenjuanbiwan group were notably higher than those of the normal group(P<0.01).The plasma TNF-α,IL-1,PGE2 contents of the Sanhuwan group and Yishen-anbiwan group were notably lower than those of the modei group(P<0.01或p<0.05),The statistical meaning of the plasma TNF-α,IL-1,PGE2 contents of the Sanhuwan group and Yishen-anbiwan group was not significant (P>0.05). Conclusion:(1)Arthritis occurenced in the three groups rats that were established by subcutaneous injection ofⅡcollogen,and that showed the model of rheumatoid arthritis was successful.(2) TNF-α,IL-1,PGE2 accelerated the formation and development of synovitis in rheumatoid arthritis, and TNF-α,IL-1 and PGE2 maybe affected and accelerated each other,and that was one of mechanisms of rheumatoid arthritis.(3) Sanhuwan could significantly decrease the content of the plasm TNF-α,IL-1,PGE2 in the synovial,and that cued Sanhuwan could prevent the synovitis and that may be one of the principles that Sanhuwan treated rheumatoid arthritis.The difference of the effect that Sanhuwan and Yishenjuanbiwan blocked TNF-αand IL-1 and PGE2 were not significant... |
