Objective: To investigate the changes of AR, GSH-Px, SOD,CAT mRNA.expressional level in diabetic microangiopathy and nervous tissue. GSH-Px, SOD and CAT are antioxidant enzymes. To interpret exceptional gene expression changes of antioxidation in cerebral vessels microenvironment and blood. To identify the relation between the changes of AR expression and hyperglycemia oxidative stress .Methods: Healthy adult male 120 SD were randomly divided into normal control group and diabetic group. The diabetic group feed with high glucose and high fat bait vessel, Diabetic rats were induced by injection of streptozotocin(STZ) intraperitoneally. The ones whose blood glucose is above 13.8 is succeed. Select 15 SD rats(blood glucose: 13.8-16.7) is diabetes group Low. Select 15 SD rats(blood glucose>16.7) is diabetes group High.Then take blood and brain tissue of their group respectively. Make pathological section to observe alteration. Total RNA was extracted from the tissue and blood using the RNA extraction kit according to the supplier's protocol.RT-PCR was performed to detect the expression of different antioxidant enzymes mRNA.Results:①pathological section: normal control group : not find obviously histopathological change. diabetes group Low and High: micrangium basal lamina thickening, endotheliocyte swelling, hyperplasia, hyalinosis, lumens narrow, tubal wall to sink, support loss, gliocyte swelling, nucelus vanish, endochylema muddy, astrocyte cloudy swelling.②Diabetic SD rat group L the mRNA expression of Cu-ZnSOD (0.55±0.06) is higher than control group (0.28±0.07) significantly (P<0.05), Diabetic SD rat group H the mRNA expression of SOD(0.59±0.06) is higher than control group significantly (P<0.05), the difference of blood Cu-ZnSOD mRNA expression between the diabetic group Low and diabetes group High did not achieve statistical significence(P>0.05). the difference of Mn-SOD mRNA expression in every group did not achieve statistical significence (P>0.05). ③Diabetic SD rat group Low the mRNA expression of blood AR (208.95±11.04) is higher than control group(0.31±0.06) significantly (P<0.05), Diabetic SD rat group High the mRNA expression of AR(0.57±0.05) is higher than control group significantly (P<0.05), There is no significant difference in AR mRNA expression between the diabetic group Low and diabetes group High(P>0.05).Diabetic SD rat group L the mRNA. expression of brain tissue AR(0.52±0.09) is higher than control group (0.29±0.06)significantly (P<0.05), Diabetic SD rat group H the mRNA. expression AR(0.52±0.06)is higher than control group significantly (P<0.05), There is no significant difference in AR mRNA expression between the diabetic group Low and diabetes group High. (P>0.05).④Diabetic SD rat group Low the mRNA expression of blood GPX (0.53±0.04) is higher than control group(0.26±0.05) significantly (P<0.05), Diabetic SD rat group High the mRNA expression of GPX (0.55±0.06) is higher than control group significantly (P<0.05), There is no significant difference in GPX mRNA expression between the diabetic group Low and diabetes group High(P>0.05).Conclusion①pathological section: normal control group : not find obviously histopathological change. diabetes group Low and High: find obviously histopathological change. Indicate micrangium nerve tissue pathological changes.②the mRNA expression of Cu-ZnSOD,GPX in diabetes group Low and High is higher than normal control group, the difference of Mn-SOD mRNA expression in every group did not achieve statistical significance. Indicate the mRNA expression of Cu-ZnSOD,GPX increased feedbackly when oxidative stress is strengthened.③There is no significant difference in the mRNA expression of Cu-ZnSOD,GPX between diabetes group low and high, Indicate blood glucose concentration didn't have effects on these factors.④the mRNA expression of AR in diabetes group is higher than control significantly,but this difference can not be observed between the diabetic group Low and diabetes group High.. the difference between the diabetic group Low and diabetes group which hinted There was no significant relationship between AR mRNA expression and blood glucose.
