Objective:To observe the influence of electro-acupuncture therapy on the mitochondrial ultrastructure and apoptosis protein of hippocampal neuron of SAMP8 mice to partly explore the curative mechnism of electro-acupuncture therapy on Alzheimer's disease(AD)from protecting mitochondria and inhibiting apoptosis.Methods:SAMP8 mice were acupunctured on "Baihui(Du20)" and "Yongquan(Kidneyl)" according to the therapeutic principle of "Tonifying kidney and activating Du meridian to reviving brain and promoting intelligence".The treatment was applied once daily and totally lasted for 21 days.The changes of study and memory abilities were observed by Morris water maze to evaluate the curative effect.The mitochondrial ultrastructures were observe by transmission electron microscope(TEM),and the expressions of apoptosis protein Caspase-3, Caspase-9 of hippocampal neuron were detect by immunohistochemistry method.Results:Compareing to normal group,the learning and memory abilities of model group were decreased,the mitochondrial ultrastructure of hippocampal neurons were damaged and the expressions of Caspase-3 and Caspase-9 were increased.While compareing to model group,the learning and memory abilities of electro-acupuncture group were promoted,the damages of mitochondrial ultrastructures were relieved, and the expressions of Caspase-3 and Caspase-9 in hippocampal neuron were significantly decreased.Conclusion:The curative mechanism of electro-acupuncture therapy on AD may be related to inhibiting the apoptosis reaction of mitochondrial route and reducing the apoptosis of hippocampal neuron by relieving the damage of mitochondrial ultrastructure and reducing the expressions of Caspase-3 and Caspase-9.
