| Objective :To observe whether the prepared Chinese Medicine of nerve-I and Dexamethasone can protect optic nerve through the investigation of expression of apoptosis-related genes (Bcl-2, Bax) in the rat retina after crushing the optic nerve with microforcepsMethod: Fifty-four healthy adult Wistar rats without eyes diseases were divided into five groups randomly : normal control group, model group , Nerve -I group ,routine dose Dexamethasone group, Nerve-I accompany low dose Dexamethasone group . All the groups crush left eyes' optic nerve except the normal control group . The treatment group give medicine after making the mould . Two weeks later get thespecimen . Retinal paraffin sections were stained by immunohistochemical method to detect the expressionof apoptosis-related genes bcl-2, bax and bcl-2/bax in the retinal ganglion cells.Results: There are significant differences in the expression of apoptosis-related genes bcl-2, bax and bcl-2/bax in the retinal ganglion cells between model group and the other groups (P<0.05) except Nerve-I group (P>0.05) . There are not significant differences between the group of drug combination group and Dexamethasone group.Conclusion: Nerve-I cannot decrease RGC cell apoptosis after optic nerve crush by increasing bcl-2 gene expression and bax gene expression in this experiment, but it can decrease RGC cell apoptosisafter the low dose Dexamethasone union use . There are not significant differences between Nerve-I accompany low dose Dexamethasone group and routine dose Dexamethasone group. Above explained that Nerve-I possibly has the curative effect synergism with the Dexamethasone . |
