| Eucommia ulmoides is a kind of specific valuable economic tree species in China,which belongs to Eucommia ulmoides family and Eucommia ulmoides genus plant.It is a residual tree species in Tertiary period glacier in geology period,and unique family and genus deciduous leaf arbor.The research in modern medicine showed that Eucommia ulmoides bark has efficiencies, such as reinforcing hepatic asthenia,enhancing bones and muscles,adjusting blood pressure, preventing ageing,antioxidation,rejecting tumor,reinforcing organism immunity and so on.In resent years,researches showed that Eucommia ulmoides bark and leaves have analogical chemical constituent and pharmacological action,but it was mentioned that Eucommia ulmoides leaves has new exploitation value due to the constituents which are different from Eucommia ulmoides bark in some documents.In addition,Eucommia ulmoides polysaccharide in Eucommia ulmoides bark can enhance organism non-specificity immune function.For example,it can inhibit the adsorption and proliferation of acquired immune deficiency virocyte adsorption and preclude infection of Acquired Immune Deficiency Syndrome(AIDS).Due to its non-side effect, Eucommia ulmoides polysaccharide can be taken for a long term.The researches on Eucommia ulmoides leaf polysaccharide restricts at content detection at home and abroad.Manifold pharmacological action and physiology function of Eucommia Ulmoides aroused abroad attention.In this papers,the Eucommia Ulmoides leaf was comprehensively compared with the Eucommia Ulmoides bark.Extraction separation of Eucommia Ulmoides leaf polysaccharide and its function of eliminating free radical were researched systemically.The anti-tumor and anticomplementary activity of Eucommia ulmoides leaf polysaccharides were researched elementarily.The result indicated that Eucommia ulmoides leaf polysaccharides had remarkable anticomplementary activity,which laid the foundation for medicinal exploiture of Eucommia ulmoides leaf.In this paper,we found out some results,as following:1.Content of chemical compositions in Eucommia Ulmoides leaf and bark is variant, especially when fractional extraction method was employed.Some polar constitutes have most obvious variance in content.And,the parts of polar constitutes exist only in Eucommia Ulmoides leaf,not founded in Eucommia Ulmoides bark.2.The optimal condition of extracting Eucommia Ulmoides leaf polysacchafide by water were as following:material/solvent ratio 1:10,extracting 120min at 90℃for 2 times.The extracting rate reached 3.713%.Use the method of Sevag to remove protein.The method of decolor was as following:use macroporous resin D4020 as decolorant,room temperature(20~25℃),pH=5.4~6.44,velocity of flow 3BV/h.3.Component of Eucommia ulmoides leaf polysaccharides was analysised with DEAE-cellulose.When we used 0.1mol/L NaCl eluting,it appeared four peaks.When we used 0.2mol/L and 0.3mol/L NaCl eluting,it appeared noly one peak respectively.The component of 0.2mol/L NaCl eluting was the highest in polysaccharides.In addition,when we used 0.0,0.4,0.6, 0.8 and 1.0mol/L NaCl eluting,it did ont appear any polysaccharides.4.Eucommia ulmoides leaf polysaccharides had the cleaning capability of hydroxy radical and radical anion.The cleaning capability of hydroxy radical was higher than that of radical anion. The cleaning capability of crude polysaccharides was higher than that of refined polysaccharides.5.There were not immediate lethal effect on mortal leukemic cell K562 and mortal nonsmall-cell lung cancer in Eucommia ulmoides polysaccharides.6.Eucommia ulmoides polysaccharides had anticomplementary activity definitely,and it presented dose-effect relationship between 0.03 and 1.67mg/mL. |
