The Eucommia belongs to the Eucommia ulmoides Oliv, and was originally recorded in《Sheng Nong's herbal classic》. Eucommia is a traditional Chinese cash crop and has prolific chemical substances, such as phenylpropanoids, iridoids, lignans etc; and has various medical efficacy, such as protecting liver, anti-bacteria and detoxification, enhancing immunity, reducing blood fat and blood pressure etc. The paper studies on the extraction, separation and purification techniques of the active ingredients in Eucommia ulmoides oliv and to acquire the best routing for the deeply process of Eucommia and deeply researches how to establish the best producing process for Eucommia1) Study on the extraction techniques in Eucommia bark. By comparing the three kinds of extraction method (the soxhlet extraction, the direct extraction and the ultrasonic extraction), the experiment result indicates that the ultrasonic extraction is the best one. It is a quick and efficient method and the best production technique has been established.2) A breakthrough method of reversed-phase high performance liquid chromatography has been established for the simultaneous detection of geniposidic acid, geniposide, chlorogenic acid and pinoresinol diglucoside in Eucommia ulmoides. Separation was achieved using a Kromasil C18 column (4.6 mm×200 mm,5μm), under the gradient elution with the mobile phase of acetonitrile and 0.1%(v/v) phosphoric acid/water, linear gradient:0-7min,7%B, 7-35min,14%B,35-45min,15%B, flow rate was 1.0 mL/min, detected at 235 nm and under 30℃. The linear ranges of 4 components were 0.1025-0.5125μg/mL (R2=0.9999),0.0925 0.4625μg/mL (R2=0.9999),0.120-0.600μg/mL (R2=0.9996),0.090-0.450μg/mL (R2=0.9998) respectively. The average recovery rate (n=5) is 96.06%,96.22%,96.12%,96.08% and with RSD of 1.39%,1.82%,1.53% and 1.59% respectively.
