| Purpose: PD is one of the commonly progressive neuro-degenerative diseases of the human being, the pathological transformation mainly is the largely death of dopamine neurons which by the SNc project to the corpus striatum. The clinical symptom mainly are stilly shacking,slowly movement,rigidity muscles and unusual posture and gait etc., the presently clinical therapy mainly using the L-dopa,the dopamine agonists, the monoamine oxidase inhibitors-B, the catechol-o-methyltransferase and the Anticholinergic drugs, but none of these medicines could make lysis to the futher expansion of patients' condition, moreover, use those medicines above in a long-term would bring about comparatively strong side effects. The therapy of surgical operation's curative effects is unsure and has many side effects. Genetic therapy is still remain in animal experimental phase. Of late years, Acupuncture and Moxibustion has been largely used in clinical more and more as an unique, safe, convenient therapy, and it's acquired cnaomparatively positive curative effects. This project is designed to research the effect of the Electroacupunc towards the revolving behavior of PD rats, GDNF and the expression of the GFR-α1, to discuss the curative effects and its functional mechanism of the Electroacupunc towards PD, to provide experimental basis for the clinical acupuncture and moxibustion.Approach: The Parkinsonian rats were modeled by stereotaxical microquantitative injection in the right striatum with 6-hydroxydopamine. The same amount of normal saline was injected into the rats of control group. After 4 weeks ,Apomorphine was injected into the rats abdominal cavity to induce rotation .The test were repeated every 7 days .Rats who had constant left rotation with a speed faster than 7r/min were selected as successful PD models. Successful PD model rats were selected for treatment and randomly divided into two groups, model group and electroacupuncture group. Acupoints of Fengfu in EA group were punctured once a day for 14 days. The therapy of immunohistochemistry is adopted to quantify the GDNF and GFR-α1 positive cells in substantia nigra,All data were expressed through SPSS11.0 software system. Results: (1) In EA group, the number of rotation after the treatment was lower than before the treatment; (2) The number of TH positive cells in SNc of model group was obviously lower than that of the normal group and sham-operation group (P < 0.01) ; (3) The number of GDNF and GFR-α1 positive cells in SNc of EA group was obviously increase than that of the model group and normal group (P < 0.05 ) .Conclusion: The electro-acupuncture Fengfu acupoint could ameliorate the revolving symptom of the PD pattern rats, reinforce the expression of the GDNF and GFR-α1 of the PD pattern rats. |
