Qufeng Antispasmodic Class Of Prescription Intervention In Allergic Asthma Mice Arg-1 Gene Regulation Research

Objective:In this study,by using immunohistochemical method and real-time quantitative PCR method to study the effects of Fengxiaofang, Dingchuantang,Sheganmahuangtang and the positive control drug dexamethasone on the expression of arginaseⅠgene in ovalbumin-challenged asthmatic mice,in order to explore the possible mechanism of Fengxiaofang,Dingchuantang,Sheganmahuangtang to asthma. Methods:119 healthy BAIB/c male mice weighted(20±2)g were randomly divided into 17 groups(n=7):normal group(group A),normal group+large dose of Fengxiaofang group(group B1),normal group+large dose of Dingchuantang group(group B2),normal group+large dose of Sheganmahuangtang group(group B3),asthmatic group(groupC),small dose of Fengxiaofang group(groupD1),medium dose of Fengxiaofang group(groupD2),large dose of Fengxiaofang group(groupD3),small dose of Dingchuantang group(groupE1),medium dose of Dingchuantang group(groupE2),large dose of Dingchuantang group(groupE3),small dose of Sheganmahuangtang group(groupF1),medium dose of Sheganmahuangtang group(groupF2),large dose of Sheganmahuangtang group(groupF3),small dose of dexamethasone group(groupG1),medium dose of dexamethasone group(groupG2),large dose of dexamethasone group(groupG3).After a week of feed,each mice was sensitized by 0.4ml antigen solution(including egg albumin 20μg and aluminum hydroxide 2mg)in the first day and was sensitized by 0.4ml antigen solution(including egg albumin10μg and aluminum hydroxide 1mg)in 7,14 days from group C to group G3.After 3 times injection,the mice were challenge with 5%egg albumin for 25 minutes every morning(one times per day for 7 day).Before being challenged every day, the mice of group C were administered with 0.9%NaCl 0.6ml,groupD1,D2, D3 were administered with 0.315g/ml,1.575g/ml,3.150g/ml Fengxiaofang 0.6ml,group E1,E2,E3 were administered with 0.345g/ml,1.725g/ml, 3.45g/ml Dingchuantang 0.6ml,groupF1,F2,F3 were administered with 0.255g/ml,1.275g/ml,2.55g/ml Sheganmahuangtang 0.6ml.group G1,G2,G3 were administered with 0.063g/ml,0.315g/ml,0.63g/ml dexamethasone 0.4ml.The all disposal of mice in group A were alike the above group's but substituing 0.9%Nacl for antigen solution.24 hours after being challenged 7 days,One parts of lungs were carried through HE,others were employed to detect the lung tissue and the tracheas RNA for arg-1 gene by real-time quantitative PCR.The data were analyzed for statistical difference using the statistical package for social science(SAS). Significance levels were established with a P value less than 0.05.Results:Fenxiaofang,Dingchuantang,Sheganmahuangtang all have a downward effect to arg-1 gene in the lungs of asthmatic mice,there into,F1 group have the best downward effect,better than E1 group,D1 and G2.The real-time quantitative PCR results show that Fenxiaofang, Dingchuantang,Sheganmahuangtang all have a downward effect to the arg-1 gene in the lungs of asthmatic mice,F1 group(small dose of Sheganmahuangtang)have the best downward effect,better than D2 group(medium dose of Fenxiaofang group),E3group(large dose of Dingchuantang)and G3 group(large dose of dexamethasone);Fenxiaofang, Sheganmahuangtang have a downward effect to the arg-1 gene in the tracheas of asthmatic mice,but,not better than that of G3 group(large dose of dexamethasone),Dingchuantang do not have any downward effect.Conclusion:Fenxiaofang,Dingchuantang,Sheganmahuangtang have some downward effect to the arg-1 gene of the asthmatic mice.Some even obtained the results better than dexamethasone.Preliminary discussion the molecular mechanisms of the Fenxiaofang,Dingchuantang, Sheganmahuangtang,what make the Chinese medicine treatment of asthma more objective and effect of volume.
