Detection Of Allergic Rhinitis Patient IL-2、IL-4mRNA Between Uighurs And Han In Xinjiang By Real-Time PCR

ObjectiveTo investigate the expression level of IL-2、IL-4mRNA in the CD4+T lymphocyte of Allergic Rhinitis patients peripheral blood between Uighurs and Han in xin jiang, comparing the role of two genes in disease.MethodsThe object of study is80patients with allergic rhinitis and80cases of healthy controls,The patients with allergic rhinitis is uighurs and han Chinese in40cases each, the corresponding healthy controls is also uighurs and han Chinese40cases in each.Cases of patients from the otolaryngology Of Chinese medicine hospital of the xinjiang uygur autonomous region, and the population of healthy controls is from the xinjiang medical university volunteer.Extracting the peripheral blood5ml as the experimental specimens.(2) Separation outside individual nucleus cell (PBMC), with immune magnetic bead method extraction CD4+T lymphocyte.(3) Use SYBR Green I real-time fluorescent quantitative polymerase chain reaction (PCR) test IL-2and IL-4mRNA genes expression level.Results:Choosing the housekeeping genes GAPDH for correction, the expression level of IL-2mRNA in the AR group is (8.66±1.92) copies/ul,the Uighurs and Han group is(8.52±2.15) copies/ul and (8.81±1.79) copies/ul in each.the expression level of IL-2mRNA in the AR control group is (11.28±1.78) copies/ul,the Uighurs and Han group is (10.31±1.92) copies/ul and (12.27±0.93) copies/ul in each. The expression level of IL-4mRNA is (10.90±2.54) copies/ul in AR group by the correction of GAPDH,the Uighurs and Han group is (10.42±2.95) copies/ul and (11.41±2.13) copies/ul in each.AR in control group is (3.07±1.18) copies/ul,the Uighurs and Han group is (2.81±1.46) copies/ul and (3.46±0.61) copies/ul in each.The expression level of IL-2mRNA has a little difference between AR patients and healthy controls,Between the Uighurs and Han group has no difference.The expression level of IL-4mRNA is in Han AR group is higher than the healthy controls,Between the Uighurs and Han AR group has no difference.ConclusionThe expression level of IL-2mRNA has significant difference between AR patients and healthy controls (P<0.05).The expression level of IL-4mRNA is in AR group is higher than the healthy controls (P<0.05).The IL-2and IL-4gene are significant in the process of the AR disease,IL-2gene has no obvious difference between the Uighurs and Han AR patients,IL-4gene has no obvious difference between the the Uighurs and Han AR patients.
