Objective:Menopausal osteoporosis is syndrome shown on women caused by thefunctional descent on both ovaries and the level of estrogen. During the metabolism oftheir bones, the bone absorptive rate is higher than formative rate. As the result, theprogression of calcium deficiency and the degeneration of bone tissue are thecharacters in this period. Clinical manifestations appear on these elder women, whichare osteosclerosis and increasing the possibility of fracture. The protophase studyshow that GNAP can raise the ovariectomized rats' serum bone gla protein(BGP), bonemineral content(BMC), bone density and insulin-like growth factor, improve osteoporosis.Recently, the values of the regulatory factors for cells are gradually known by thescholars in the study of osteoporosis. They have important regulations in the bonetransition. The approach used here is based on the former fundamental research. Byusing the molecular biology technique, the cytokine system was tested to study themechanism of the GNAP treatment to the menopausal osteoporosis. By conductingexperiment, effects of defferent treatments were compared to see whether thedifference exists, and provide theoretical evident on clinical manifestation prophylaxisand treatment.Methods:In this experiment, 30 rats which is between eleven-month-old andfifteen-month-old were chosen as the experimental objects, and their weights areranged from 300 to 400 grams. The rats were randomly classified into three groups,10 rats in each group. They were GNAP treatment group, estrogen treatment groupand the menopausal group. 10 rats which is between four-month-old andsix-month-old were chosen as the youth reference group, and their weights are rangedfrom 130 to 180 grams. All medicine treatment groups were treated for four weeks,both the menopausal group and the youth group were drenched Sodium Chloride forfour weeks which equal to the amount that the medicine treatment groups were given.Four weeks later, all rats were excised eyeballs to take blood, the blood wascentrifugated to test IL-6 by RIA(Radioimmunoassay), and all humerus were takento weigh their weight(the dry weight and the wet weight) and measured their lengthsand width, and their bone strengths were simultaneously detected.Results:1. IL-6 of the menopausal group was obviously higher than that of the youth group, and the bone strengths were lower (P<0.01).2. After the treatment of GNAP and extrogen, IL-6 was obviously decreased(P<0.05), and the bone strengths were obviously increased(P<0.01), but the therapeuticefficacy of the GNAT treatment group and that of the estrogen treatment group had noobviously difference(P>0.05).3. Both GNAP and estrogen can decrease IL-6 and increase the bone strengths,weights, widths and lengths. The therapeutic efficacy had no obviously difference(P>0.05).Conclusion:GNAP can decrease IL-6 of blood serum, and raise bone strengths, weights,widths and lengths, decrease the bone rebuilt, which decreased the activities of thesecrete cells, and the activation and quantity were decreased accordingly, the boneresorptions were inhibited ultimatedly. Meanwhile, GNAP probably increased theactivities of the osteoblast, and raised the bone mass. The mechanism of action ofGNAP and that of estrogen was difference. GNAP can both inhibited bone resorptionand promote bone formation, which increased the bone density of the menopausalrats, the purpose of treat osteoporosis was reached. The new theory on clinicaltreatment was provided.
