ObjectiveHuangjiao-Granule is the unique praeparatum for stroke treat in acute stage.Clincial observation confirmed its therapeutic effect,animal study show that it can improve hemorheology and antagonism apoptosis ,this research from the exvivo cell level investigate the protective effect of the phamacological serum of HuangJiao Granule on neuronal cell (SH-SY5Y) induced by hypoxia-ischemia injury; Search the optimal concentration for the neuron protective effect, Explore the mechanism of the protective effect on neuronal cell following hypoxia-ischemia under the optimal concentration ,identify the inhibit effect on neuron apoptosis following hypoxia-ischemia.Methods1 After grouping animal,administration by gavage,blood sampling,made HuangJiao Granule phamacological serum and blank controlled serum.2 Use sequencly cultured neuronal cell SH-SY5Y, Copy neuronal cell injury modle induced by hypoxia-ischemia culture for 15 min, intervene neuronal cell with phamacological serum,blank controlled serum and differently concentration phamacological serum.3 By the method of MTT microcolorimetry to detect the neuron activity of diffetent serum and of different concerntration.4 Detect cell apoptosis rate by flow cytometry method under the optimal concentration.Result1 Hypoxia-ischemia culture leads to cell injury: under the same serum condition ,the activity of neuron following hypoxia-ischemia culture decreased distinctly in comparsion with the normal culture (P<0.01).2 The neuron protective effect of HuangJiao Granule:at the same serum comcntration of 10%, the neuron death rate of HuangJiao Granule group is siginificant reduced compared the blank controlled group (P<0.01),which show the neuron protective effect, death rate＝(ODnormal - OD_HI ) / ODnormal3 The comparison of neuron protective effect in different concentration: in the different concentration group of 5%,10%,20%,40%, the neuron protective effect of group 20% and 40% is significant preceded (P0.05)4 The mechanism of the protective effect: detected by FCM show the way of neuron death induced by hypoxia-ischemia including necrotic and apoptosis , apoptosis is the important format, the mechanism of the protective effect ofhuangjiao granule on neuronal cell is reduced neuron apoptosis compared wjth blank serum.Conclusion1 The phamacological serum of HuangJiao Granule has actual neuron protective effect.2 The mechanism of the protective effect of huangjiao granule is reduce neuron apoptosis.
