Methotrexate And Cyclophosphamide In Rat Cell Dna Damage

Objective To compare the effectiveness of methotrexate(MTX),cyclophosphamide(CTX) and combination of two drugs to rat's different cell.To authenticate that specificity of drug action could be elevated by combination.Method To detect DNA damage of rat's bone marrow cell,spleen cell,peripheral blood lymphocyte,hepatic cell,renal cell and sexual cell induced by MTX,CTX and combination of two drugs.DNA damage is evaluated by single cell gel electrophoresis(SCGE).DNA damage is estimated by DNA migration and comet frequency.Result1.After treatment of MTXDNA migration and comet frequency of peripheral blood lymphocyte,hepatic cell,renal cell and sexual cell are not different from the control group significant.DNA migration of bone marrow cell and spleen cell are different from the control group significant.Comet frequency of spleen cell is different from the control group significant.2.After treatment of CTXDNA migration and comet frequency of renal cell is not different from the control group significant.DNA migration and comet frequency of bone marrow cell,spleen cell,peripheral blood lymphocyte,hepatic cell and sexual cell are different from the control group significant and were exceed the effect of MTX.3.After treatment of combinationDNA migration and comet frequency of peripheral blood lymphocyte,bone marrow cell, spleen cell,hepatic cell and sexual cell are different from the control group significant.DNA migration of renal cell is not different from the control group significant and comet frequency of it is increased.DNA migration of bone marrow cell,spleen cell and peripheral blood lymphocyte are increased significant contrast to using CTX only.Comet frequency of spleen cell, peripheral blood lymphocyte and renal cell are increased significant contrast to using CTX only. DNA migration of renal cell and comet frequency of bone marrow cell are not different from using CTX only significant.DNA migration and comet frequency of hepatic cell and sexual cell are not different from using CTX only significant.Conclusion Effectiveness of MTX and CTX to different cell is unequal.Effectiveness of CTX is stronger than MTX.Specificity of CTX is less than MTX.Combination of MTX and CTX could increase the dugs specificity.
