Radix Platycodi is the root of P. grandiflorum(Jacq.)A.DC., family Campanulaceae (< Pharmacopoeia of P.R.C>, 2005 edition ) , It shows the effects of dispersing the lung to resolve phlegm, soothing the throat, and draining the pus. P. grandiflorum is frequently used traditional Chinese drug clinically. The quality controlling methods merely are conventional character identification, TLC identification and weighting method is used to determine contents of the saponins. The quality criteria and processing technology of processed product are far away from normalization. To ensure therapeutic effect of P. grandiflorum, this paper determined the contents of platycodin D, instituted quality criterion for < Pharmacopoeia of P.R.C>, 2010 edition, and instituted quality criterion and processing technology of processed product. This paper approached the methods of processing in the planting area and isolated chemical constituents from the aerial parts of P. grandiflorum.This paper includes three parts:Part one: This paper has mainly documents about the progress of P. grandiflorum on the medicinal plant resources, processing methods, chemical constituents, quality standard, pharmacological effects and clinical application in recent ten years, which has provided the important references for its further study, exploitation and application.Part two: The quality criterion on ten batches of medical material of P. grandiflorum were instituted in this paper. Platycodin D was used as reference substance in TLC qualitation identification, HPLC fingerprinting quantitation identification and HPLC quantitation determination. Meanwhile the medical material of P. grandiflorum were detected by ash content, moisture content and its extract. Making out a draft of quality criterion of the medical material of P. grandiflorum ( Draft). The quality criterion on ten batches processed product of P. grandiflorum were instituted in this paper. Platycodin D was used as reference substance in TLC qualitation identification, HPLC fingerprinting quantitation identification and HPLC quantitation identification. And the processed product of P. grandiflorum were detected by ash content, moisture content and its extract. Combine the practical condition of manufacturer, and refer to the traditional evaluation criterion. This paper verified the normalization research of P. grandiflorum's processed product and determined production technology. Part three: The methods of processing in the planting area, cheminal constituents from the aerial parts of P. grandiflorum and comparative study of several regionals of P. grandiflorum. This paper has researched on the methods of processing in the planting area and reached the conclusion that it is better not to cut the rootlet and root cortex. This paper has determined the Platycodin D and saponins of the whole plant and also determined the HPLC fingerprint of the stem and leaves. There are 5 compounds have been isolated with silica gel column chromatography. On the base of their spectral data, 4 compounds were elucidated as acacetin, luteolin, acacetin-7-O-rutinoside and luteolin-7-O-β-D-glucoside. By the direct comparison of the HPLC, the highest two peaks were identified as acacetin-7-O-rutinoside and luteolin-7-O-β-D-glucoside in the HPLC fingerprint spectrum of stem and leaves.In short, Platycodin D, which is the main active component was used as reference substance when instituted quality criterion of P. grandiflorum ( Draft ). The draft can be provided to < Pharmacopoeia of P.R.C>, 2010 edition. Meanwhile, standardized processing technology of the processed product. This paper has researched on the methods of processing in the planting area, the chemical constituents of the aerial parts and comparative study of several regionals.
