Metastasis, which is the essential character of biological behaviors in malignancies, is also the main reason for the death of tumor sufferers. So it is very important to study tumor metastasis for the prevention and cure of carcinomas. Chinese medicine considers that blood stasis symptom ties up with tumors and the therapy of blood-activating and stasis-dissolving can prevent the blood-metastasis of malignancies not only by improving blood high-coagulate state of tumor sufferers, preventing tumor emboli, but also by killing directly or indirectly tumor cells in blood circulation which break away from original tumor. However, some researchers believe that medicines for blood-activating and stasis-dissolving are profitless for tumor cure because of the facilitating effect on the metastasis of tumors. In this paper, we studied the effect of representative traditional Chinese medicines for blood-activating and stasis-dissolving (Danggui, Danshen, Ezhu, Jixueteng, Honghua, Tubiechong) on metastasis-associated capacities of B16-BL6 metastatic mouse melanoma cells so as to discuss preliminarily the relationship between blood-activating and stasis-dissolving method and tumor invasion and metastasis.The main results were as follows:1. The cytotoxicity of medicines to B16-BL6 cells was evaluated by MTT assay and cck-8 assay. The results showed that all the traditional Chinese medicines we mentioned above had an effect on the proliferation of B16-BL6 cells, but the cytotoxicity of all medicine extracts was not very great.2. The effect of medicines on adhesion of B16-BL6 cells was determined by cell-Matrigel adhesion assay. The results showed that three of the medicines exerted distinctly repressive effect on adhesion of B16-BL6 cells to LN except that Danshen and Honghua exhibited prominent facilitation while Danggui behaved bidirectional adjustment (repressed adhesion at high concentration while stimulated adhesion at low concentration).3. Invasive capacity and migrative capacity of B16-BL6 cells were tested by Transwell Chamber assay. The results showed that five of the medicines exerted distinctly inhibitory effect on invasive capacity of B16-BL6 cells into reconstituted basement membrane while Danggui had unobviously repressive effect on invasion of B16-BL6 cells.
