Objective: To explore the relationship oxidative stress/lipid peroxidation and nonalcoholic steatohepatitis , and to evaluate the effects of the Chinese herbal compound Jiangzhiyiganchongji on nonalcoholic steatohepatitis in rats.Methods: Forty-eight male Wistar rats were randomly allocated into 6 groups. Control groupâ… ,â…¡were fed with general diet and model groupâ… ,â…¡were fed with high fat diet.Medication rats were given Jiangzhiyiganchongji orally after fed with high fat food 8 and 12 weeks.After 12 and 16 weeks,the rats were sacrificed respectively.Bodyweight,body length,liver weight, serum lipid content, live constitution homogenate MDA,FFA level and GSH-PX activity were measured quantitively and liver histology were detected.Results: After 12weeks,model group rats developed lipid metabolism disorder and all the model rats presented the patho1ogy of modern-severe hepatocyte steatosis, inflammatory infiltration and necrosis in livers.And every marker of administration group was improved significantly.After 16weeks the level of live constitution homogenate MDA went towards the decreasing tendency. The level of FFA were significantly higher than that of model groupâ… . The hepatic GSH-PX activity is lower but there is no significant difference when compared with model groupâ… . In administration groupâ…¡the level of MDA , FFA decreased significantly and GSH-PX activity improved prominently. But MDA level was still higher than that of control groupâ…¡unsatisfied. Liver histology showed obvious improvement in steatosis and inflammation.Conclusions: (1) Oxidative stress/lipid peroxidation play a key role in the formation and development of liver inflammation. (2) FFA may be crucial in the pathogenesis of NASH and linked closely with lipid peroxidation. (3) Jiangzhiyiganchongji is approved to be operative in prevention and cure nonalcoholic steatohepatitis by improving the anti-lipid peroxidation capability and reducing the FFA levels. |
