Patients With Preeclampsia Bfgf With Nf-kappa B, Sflt-1 And Its Relationship,

Objective To investigate the change of soluble fms - like tyrosine kinase 1 (sFLT - 1), basic fibroblast growth factor(bFGF) and the activity of nuclear factor kappa B(NF- KB) in the hypertensive disorder complicating pregnancy(HDCP) , verify the relationship of them in HDCP's development and progression and reveal the effect of them. Method ①In suit hybridization was adopted to test expression level of NF - KB mRNA in placenta of Hypertensive disorder complicating pregnancy (including 23 gestational hypertension, 20 mild preeclampsia,36 severe preeclampsia) and 30 normal pregnant women. ②The expression of NF- KB,bFGF protein in placental were measured by immunohistochemistry streptavidin -biotin peroxidase(SP) method. ③Enzyme - lingked immunosorbent assays (ELISA) were used to determine pregnant women serum and the umbilical serum level of bFGF and soluble fms - like tyrosine kinase 1 (sFLT - 1). Result ①The positive expressions of NF - KB mRNA and protein were all located on cytoplasm, the expression tendency of them was consistent, The level of NF - KB mRNA increased with the seriousness of disease. The level of NF— KB mRNA in preeclampsia were siginificantly higher than normal pregnancy(P<0. 01). severe preeclampsia were higher than mild preeclampsia (P< 0. 05) and mild preeclampsia were significantly higher than gestational hypertension(P<0.05) . gestational hypertension were significantly higher than normal pregnancy(P<0.05). The expression of NF - KB protein were consistant with the level of mRNA. ②The expression of bFGF located in placental syncytiotrophoblast,cytotrphoblast,decidual cells and endothelial cells plasma. In preeclampsia group were significantly higher than that in normal pregnancy group(P<0. 01).
