ObjectiveTo explore the etiology and pathology of depression beginning with depressive disorder of TCM. To observe the effects of Pill for Relieving stagnancy to CUMS mouses for analyzing the mechanism of action of Pill for Relieving stagnancy for depression.Methods1.Grouping: 40 mouses were randomly divided into 4 groups according to body weight: normal group, model group,control group and herb group of Pill for Relieving Stagnancy ,12 mouses per groups.2.Modeling: to establish CUMS mouse model referring to literature except to the normal group.3.Giving medicine:After 18 days from modeling, Decoctum of Pill for Relieving Stagnancy was given to the herb group, 0.1ml/10g,the density of the decoctum was 13g/ (kg · d) . Amitriptyline was given to the control group. Equal volume of isotonic Na chloride was given to the normal group and model group.4.Detection: To record the action information of mouses with BL-410 organism function experiment system.And compare the contents of 5-HT, hematoplasma hydrocortisone.Results1. To compare with the model group, the autonomic activities of herb group of Pill for Relieving Stagnancy and control group increased significantly (p<0.01). There were no significant differences between the herb group and normal group (p>0.05 ). And there were no significant differences between the herb group and control group (p>0.05 ) .2.The irritation of herb group and control group significantly decreased (p<0.01) . There were no significant differences between the herb group and normal group(p>0.05 ) ,but There were significant differences between the herb group, normal group and control group (p<0.01) .3.To compare with the normol group,the contents of 5-HT of herb group significantly decreased (p<0.01) . To compare with model group, the contents of 5-HT of herb group and control group significantly increased (p<0.01) . There were no significant differences between the herb group and control group (p>0.05) .4. The density of hydrocortisone in blood of the herb group and control group significantly decreased compared with model group(p<0.01). There were no significant differences between the herb group and control group (p>0.05) ,and there were no significant differences among the herb group, normal group and control group (p>0.05) .ConclusionHepatic Qi stasis and emotions oppression is a very important link which causing depression. Pill for Relieving Stagnancy can effectively improve the CUMS mouse's depressive symptom.The mechanism is possibly related to that Pill for Relieving Stagnancy can increasing 5-HT and decreasing the content of hydrocortisone in blood.
