| Objective:To observe levels of plasma endothelin (ET),calcitonin gene-related peptide(CGRP) and brain natriuretic peptide(BNP) in rats with myocardial hypertrophic heart failure affected by BaoXinHeJi,explore the mechanism and offer the scientific basis for heart failure curing of TCM. Methods:56 healthy Wiistar rats were choosed and randomly divided into seven groups: normal control group, model group A, model group B, model group C, high dosage of BaoXinHeJi group , low dosage of BaoXinHeJi group. Except the rats of normal control group, others were subcutaneous injected isopenaline for 13 days,with first dosage of 20, 10 , 5mg/kg in 3days and then dosage of 3 mg/kg in 10 days. BaoXinHeJi was given with high dosage 2ml and low dosage 2ml, bid, when making models. The Digoxin was given with the dosage of 0.27 mg/kg,qd. The normal control group was given 0.9% sodium chloride solution 2ml,bid. The treatment period of all groups was 13 days.After making models (giving drug was over at the same time), all rats except rats of model group Band C were tested plasma ET, CGRP by radioimmunoassay method, and BNP by ELISA method, then were killed and weighed heart , lung and liver. From the fourteenth day, the rats of model group B and C were given high dosage of BaoXinHeJi 2ml,bid and 0.9% sodium chloride solution 2ml, bid respectively. The period of treatment was 14 days.After that all rats were tested plasma BNP by ELISA method and then well killed and weighed heart , lung and liver. Result:1. ET tested result: The normal control group:(310.24±41.72) ng/1, the model group A:(472.74 ± 38.37) ng/1, The high dosage of BaoXinHeJi group:(237.87± 26.22) ng/1, The low dosage of BaoXinHeJi group:(305.82 ±22.48) ng/1, The Digaoxin group:(310.24 ±41.72) ng/1. The level in model group A was higher significantly than that in normal control group( P<0.01). The high dosage and low dosage of BaoXinHeJi could decrease ET level sharply (P<0.01), and obvious difference was not seen among digaoxin group and them (P>0.05).2. CGRP tested result : The plasma levels of CGRP in nrmal control group ,model group A, high dosage of BaoXinHeJi group, low dosage of BaoXinHeJi group ,and digaoxin group were (287.32 ±44.78) ng/1, (176.22 ±18.23) ng/1, (240.23±36.51) ng/1, (181.23±14.28) ng/1, and (278.83± 44.23) ng/1 respectively. The level in model group A was lower significantly than that in normal control group( P<0.01). The high dosage of BaoXinHeJi could increase CGRP level strikingly(P<0.01), and there had not obviousdifference compared with digoxin group (P>0.05).3. The first BNP tested result: The plasma levels of BNP in the five group as described about ET and CGRP were (0.272 ± 0.072) ng/ml, (1.322 ± 0.272) ng/ml, (0.131±0.129) ng/ml, (0.718 ± 0.097) ng/ml, and (0.310 ± 0.066) ng/ml respectively. Comparing with normal control group, The level in model group A was higher (P<0.01). The high dosage and low dosage of BaoXinHeJi could decrease BNP level significantly (P<0.01) , and there had not obvious difference among digoxin group and them (P>0.05).4. The second BNP tested result : The second BNP testing showed BaoXinHeJi high dosage had good curing effect on heart failure. The level in model group B(0.494 ± 0.038 ng/ml) and model group C(0.952 ± 0.354 ng/ml) had significant difference (P<0.01).5. Weighing heart, lung and liver for the first time :The level in model group Aand normal control group had significant difference (P<0.01). High dosage of BaoXinHeJi and digoxin could decrease weight of heart, lung and liver obviously (P<0.01).6. Weighing heart , lung and liver for the second time :The levels of lung andliver model group B were lower than those in model group C (P<0.05). The weight of heart had not obvious chang in both groups. Conclusion:BaoXinHeJi has good preventing and curing effect on rats with myocardial hypertrophic heart failure as well as digoxin, especially high dosage. From BaoXinHeJi could adjust ET, CGRP and BNP levels, we infer its mechanism maybe improve neuroendocrine... |
