Objective: To study the relationship between the platelet activation andthe disease activity of rheumatoid arthritis(RA)by measuring the expression onplatelet surface of CD41/CD61, CD62P in patients with RA,explore the effect ofplatelet activation in the cardiovascular disease of RA patients. Methods:Flow cytometric assay of platelet-rich plasma ,two-colour technique, MoAb FITI,PE-conjugated was used in 43 patients RA and 20healthy individuals. Result: The positive rate of CD62P,PLT and ESR in active group (23patients)( ( 27.8± 8.5 ) %,(278.6± 84.2) × 10~9/L, (51.1± 36.3) mm/h,respectively) were higher than those in remission group (20 patients)(13.6±3.9)%, (189.6± 64.6) ×10~9/L , (8.5± 4.2) mm/h ,respectively),P<0.05.On the other hand, 9 patients with thrombocytosis showed the increased levelsof WBC,ESR ( (8.3± 2.3)×10~9/L, (51.4± 39.1) mm/h, respectively) which werehigher than those in 34 patients with normal thrombocyte( (6.9± 1.5) ×10~9/L ,(25.9 ±31.1)mm/h , respectively),P<0.05. There was no difference of CD62Pexpression between thrombocytosis group and normal thrombocytegroup((32.2± 8.7)% vs(18.1± 6.3)%,respectively).The expression of CD62Pand WBC in RA group( 43 patients) ((20.8± 7.0)%,(7.2± 1.8)×10~9/L,respectively) were higher than those in healthy group (20 patients) ((7.9 2.0)%,(5.7± 1.0)×10~9/L , respectively),P<0.001.There were no difference in theexpressions of CD41, CD61 between each group. There were not relationsbetween the expressions of CD41, CD61, CD62P and DAS, WBC, PLT, ESR,P>0.05. Conclusion:Present study indicates that the high expression of CD62P onplatelet surface is related to the disease activity of RA. Platelet may have aneffect on the inflammation progress of RA by releasing P-selectin afteractivation. The high expression of CD62P may be the one of the mechanismsthat lead to cardiovascular disease of patients with RA. The CD41/CD61 mayhave no relationship with the disease activity and cardiovascular disease ofpatients with RA.
