Objective:To establish a method for detection of platelet microparticle (PMP), glycoprotein IIb/IIIa (PAC-1) and P-selectin (CD62P) using whole blood flow cytometry(WB-FCM) through comparison to the detecting approach of platelet activating flow cytometry (PA-FCM) that was performed for control; To explore the effects of different agents for platelet activation and different sample treatment approaches on the detecting results of PMPs, GPIIb/IIIa (PAC-1) expression and P-selectin (CD62P) expression; To further elucidate the relationship between the quantative changes of PMPs in whole blood before and after the treatment in patients with ischemic cardiovascular and cerebrovascular diseases and the activation ratio of PAC-1 and CD62P based on the experiment observation; To explore the role of PMPs, PAC-1 and CD62P in the pathogenesis of thrombotic cardiovascular and cerebrovascular diseases as well as the clinical significance of detecting PMPs and activation ratio of PAC-1 and CD62P in whole blood in the prediction of illness state and in the prognostic evaluation.Methods:1.
