| Objective It was reported that rALR (rat augmenter of liver regeneration) distribute in most of tissue and organ of rat, no-tissue specility; mRNA expression of rALR was identified variedly in different tissue, strongest in testis. But nowadays there was not some study on ALR in rat testis. Here to confirm the expression and localization of ALR in infancy,adult and old rat testis,comparing the change of ALR expression degree before per 70 PH and after per 70 PH. In order to evaluate expression principle of ALR in rat testis and know if ALR has tight connection with spermatogenesis and development of testis, to explore the relation between ALR and reproduction, to discover the possible expression character and functional methods of ALR. Methods Infancy(who age are 1 week~1 month), adult and old rat were respectively divided into three groups , the number of each group was eight, whose testis were resected under anesthesia with ether , by which adult rat testis and liver were resected in no-surgical group, in 12h, 1 day, 2 days and 3 days after per 70 PH, each groups had 5 rats. Testis were fixed for 2 days with bouin`s liquor, liver issue were immobilized with per 10 formaldehyde solution for 1 day, Stabilizated by paraffin and sected. Expression and localization of ALR were examined by S-P immunohistochemistry. Result Expressing in cytoplast and membrance of cell, ALR exist in Leydig cells,sperm cell and Sertoli cell of rat testis, especially stronger in spermatogonia and spermatocytes. ALR expression vary significantly in infancy, adult and old rat testis, which level were respective strongest, stronger and weak (P<0.05). The expression of ALR in liver became stronger after per 70 PH, peaking in 24h, but which didn`t change in testis. Conclusion In testis ALR expression is strong which is necessarily for testis development and were connectede with spermatogenesis. The level of ALR in different organ is different. The function of ALR is special for different organ. |
