Objective:The climacteric is the transitional period of a woman from bearing to agedness. During this period, ovarian atrophy and dysfunction gradually, that excretion of estradiol(E2) decreased markedly. As result, the hypothalamus-pituitary-ovarian axis balance was broken down. Part of women in this period can not adapt to this change and may have many unhappy feelings such as hot, flashes, nervousness, depression,> irritabilit, insomnia and so on, namely the Climacteric Syndrome. According to Bian Zheng Lun Zhi of TCM, most of this disease attribute to kidney deficiency(especially kidney-yin deficiency,79% in this disease). Liu-wei-di-huang soup is a typical traditional compuound of growing kidney-yin. It has effects of adjusting immunity, anti-tumor, lowering blood-Glu and blood-lipid, protecting liver and adjusting function of kidney according to modem pharmacology experiment. Regulating the balance of nerve-incretion-immunity network is its main pharmacology effect, and this is modern pharmacology foundation of its growing kidney-yin. In this experiment we will probe into effect of liu-wei-di-huang soup on reproductive endocrine function and target organ in ovariectomized rats, and clarify mechanism of effect of liu-wei-di-huang soup on climacteric syndrome, and give some evidence for popularizing clinic application. Method:30 female Wistar rats of adult and un-pregnant(body weight210-250g), were devided randomly into three groups(model group,liu-wei-di-huang soup group, sham operated group). Rats in model group and liu-wei-di-huang soup group were removed their ovaries. Three groups were treated with correspond treatment. After 30 days, extracting rats' eyeballs to collect blood, separating serum, determining content of E2, FSH and LH in serum by Radioimmunoassay method. Smearing vagina mucosa cell, HE staining, cytology examination. Removing uterus, fixation, embedding, section and HE staining for histology examination. Result:1. The change of serum sex hormone after being emasculated. The content of serum E2 in model group was lower than that of sham operated group(p<0.01). The content of serum FSH and LH in model group was higher than sham operated group(p<0.01,p<0.05).2. The change of serum sex hormone after 30 days: The content of serum E2 in liu-wei-di-huang soup group was higher than that of model group(p<0.01). The content of serum FSH in liu-wei-di-huang soup group was lower than that of model group(p<0.05). There was no significant difference between serum LH in liu-wei-di-huang soup group and that of model group(p>0.05).3. The change of vagina mucosa cell smear: The content of estrogen in model group was significantly lower than that of sham operated group. The content of estrogen in model group was significantly lower than that of liu-wei-di-huang soup group.4. The change of rats uterus tectology : The uterus endometrium in model group was thinner than sham operated group, it is only 1/3 of uterus wall, and secretory material decrease in epithelium, uterus gland of basic-layer decrease, gland cavity decrease. The uterus endometrium in liu-wei-di-huang soup group was thicker than model group, and it is 1/3-2/3 of uterus wall. Secretory material increase in epithelium, uterus gland of basic-layer increase. There are abundant blood vessel. Conclusion:The results above indicate that liu-wei-di-huang soup can enhance content of serumE2 in ovariectomized rats, lower FSH, add keratinocytes cells and basic-layer cells in vagina mocucity epithelium, thicken uterine endometrial, improve the quantity of gland, delay the atrophy of uterus in ovariectomized rats, and so it can regulate gonad axis on whole.
