Syndrome, which bases on the theory of Zang and its outer signs in Chinese medicine and instructs the clinical diagnosis and treatment, has been paid expansive attention in studying of traditional Chinese medicine in recent years. As basic syndrome of liver diseases of Chinese medicine is concerned, liver stagnation syndrome is one of common syndromes in clinical practice and highly valuable for profound research. Some researches indicate that liver stagnation syndrome is likely to result from regulating function disorder of high nervous centre induced by negative psychic stress in which pathological changes of multi-system including nerve, endocrine, circulation, digestion, immunity, feeling and motion etc are involved. As a complicated syndrome, it is not only liver disorder itself but also complex system of multiple Zang-fu and organs of total body are involved in liver stagnation syndrome. However, only part of known gene expression products (receptor, enzyme, cytokine and peptides etc) change is disclosed so far in some aspects, essence of liver stagnation syndrome is far away from uncovering. Complex manifestations of syndrome are likely to have some material basis which is most likely to originate from gene modulating disorder to its core. As above-noted knowledge is concerned, the following aspects were primarily explored in this thesis.Theoretical researchAs the advent of era of post-genome, focal point of studying in life science has been transformed from genome to function genome. This doctrine which pays more attention to gene function and gives more emphasis on interaction and integration is in accordance with academic characteristics of Traditional Chinese Medicine. As syndrome research is considered as importance and crucial issue of development of traditional Chinese medicine, presenting the notion of syndrome genome is urgently needed to cater for this chance and tendency. Syndrome genomics is that method of function genome is applied to discover all the genes and their functions related with some syndrome and make clear its essence in total gene expression through exploring the gene variation and differential expression of syndrome in which same disease is divided into different syndromes and different diseases are classified as same syndrome when theory of syndrome is taken as leading principle. Subject of syndrome of Chinese medicine is likely to be rapidly enhanced and to step into a high level by profound research about the new subject which syndrome is combined and integrated with genome. Therefore, presenting the notion of syndrome genome is necessary and inevitable for profound advancement of subject of syndrome in Chinese medicine.Experimental researchaLivaer stagnation syndrome model of chronic unpredictable stress was copied and hippocampus was taken to analyze the differential expression of mRNA in order to find gene segments closely related with liver stagnation syndrome and explore its material bases of its gene change for studying its essence and mechanism.Male SD rats were used for animal experiment and divided into two batches, which the first was for differential expression of gene and the second for transmembrane of Northern blot. Rats born in the same roost were chosen to eliminate individual distinction as far as possible. 6 male SD rats (200 - 250g each) were randomly divided into the normal control group which 2 rats was put in the same cage, the group of stress model and the group of stress model administered with amitriptyline which each rat was fostered solely in the first batch. In the second batch, 32 rats were randomly divided into four groups and 8 rats in each group which way of class, feeding, stimulating and administration in the first three groups was the same as in the first batch and the fourth group was model group administered with Xiaoyaosan and each rat was fostered solely.
