Viral myocaditis is the frequent disease caused by virus. The incidence is rising year after year and it is affirmed that it can cause dilated cardiomyopathy. So the medication of viral myocarditis comes to front. This article was discusses the effect of anti-virus and protecting coxsackie B3 virus induced myocardium cells by folium isatidis. It also provides the clinical application the theoretical basis of therapy. This experiment was made the model of myocaditis by inoculating coxsackie B3 virus in newborn rats?myocardium cells with the concentration of 100TCID5O. The experiment is set up 8 groups group A: healthy group; group B: inoculation the coxsackie B3 virus and folium isatidis solution at the same time; group C: inoculation the coxsackie B3 virus and carbamidine hydrochloric at the same time; group D: adsorb the cells for 2hours then washed it out and add folium isatidis solution; group E: adsorb the cells for 2hours then wash it out and add carbamidine hydrochloric; group F: cultivate the cells in folium isatidis solution for 24hours then washed it out and inoculate virus ; group G:cultivate the cells in carbamidine hydrochloric for 24hours then wash it out and inoculate virus: group H: virus-infected group. The experiment determines the virulence and the action of inhibition in virus-infected SD rats?myocardium cells cultured by CH and folium isatidis through MTT method. Observe Morphology of myocardium cells, pulsation, and ultra-structure changes through light microscopy and electronic 3 microscopy. Measured the cellular LDH (Lactate Dehydrogenises LDH) , GOT (Glutamate Oxal-acetate Transaminase GOT) through colorimetric method. Determine actibation of SOD (Super oxide Dismutase SOD) and content of MDA (Maldondialdehyde MDA) through biochemical detection. Discuss the function of folium isatidis to the CVB3-RNA by in situ hybridization method between virus-infected groups and medicine groups. Observe the function of folium isatidis resisting the coxsackie B3 virus induced apoptosis by TUNEL method. Results: (1) through the light microscopy and electronic microscopy抯 observation,it is indicated that folium isatidis canefficaciously significantly inhibited the pathological changes of Morphology of myocardium cells, pulsation, and ultra-structure. (2) Through MTT method,it is indicated that folium isatidis can raise the livability of CVB3 induced myocardium cells. (3) Through biochemical detection and histochemistry staining it is showed that folium isatidis can significantly reduced the content of LDH GOT MDA released by myocardium cells and increasethe activation of SOD strikingly (4) AS a result of in situ hybridization method it is showed that folium isatidis can significantly reduce the area of positive signal. (5) Through the TUNEL method,it is showed that folium isatidis can significantly reduce the number of apoptosis. The results of experiment showe the folium isatidis has the function of anti-virus action and protecting heart muscle cells from CVB3-induced myocarditis in SD rats.
