[ObjectivelCoxsackievirus B3 is one of the most important pathogens which can cause viral myocarditis(VMC), but the pathogenesis of VMC has not been elucidated until now. Most researchers consider that virus can directly result in myocardial lesions in the early stage of VMC, but the important pathogen is immunopathological destruction caused by virus and damaged myocardium in later stage of VMC. No method is effective on treating VMC until now, and most studies admit that Chinese Western integrated treatment is a new pathway. The objective of this experiment was to investigate therapeutic effect and mechanism with Astragalus membaranceus > Folium Isatidis and Chinese Yam on murine viral myocarditis by CVB3.-4-[Methods]Balb/C mice were infected (O.lml intraperitoneally) with CVB3 diluted in saline, and then were divided into four groups: natural saline were perfused into murine stomach in the infected control group; Astragalus membaranceus solution, Folium Isatidis solution and Chinese Yam solution were given respectively using the same approach in the rest mice. Mice were killed on day 3 ^ 5> 7^ 1(K 14> 21 after infection with CVB3 in order to observe myocardial lesions and estimate myocardial histopathological scores. Virus was isolated from murine myocardium. Perform expression levels in infiltrating cells in murine myocardium were determined by immunohistochemistry. To visualize the ultrastructure of the cardiac myocyte, heart ventricles of mice sacrificed on day 7 post-infection, which is the time when myocardial inflammation reaches maximum severity, were fixed in 4 percent glutaraldehyde.[Results]1 Light microscope of normal cardiac myocytes show that its structural arrangement is clearly; its nucleolus is obvious and clarity; its cytoplasm is enrichment; its membrane is integrity and there were no infiltrating cells among cardiac fibrin. Infiltrating cells were discovered on day 5 post-inoculation, and mass cell infiltration and cardial myocyte necrosis were detected on day 7 post-infection in infected control group. The most significant damage of myocardium were displayed from day 7 to 10 post-infection. Compared with myocardial histopathological scores of infected group, that of both Astragalus membranaceustreated group from day 5 to day 21 post-inoculated and Folium Isatidis treated group on day 5 and day 7 post-inoculated showed significant decrease (P<0.05) , but the scores had not significant difference among infected control group ^ Folium Isatidis treated group from day 10 to day 21 post-inoculated and Chinese Yam treated group from day 3 to day 21 post-inoculated.2 Electron microscope of ultrathin section of myocardium in infected control mice revealed mitochondrial swelling, mitochondrial crista rupture, sarcoplasmic reticulum dilatation and vascular endotheliocyte edema. Compared with the pathological changes of infected control murine myocardium, that of cardiac myocytes in treated groups were significantly diminished, especially that of Astragalus membranaceus treated group. There were a lot of lysosomes in myocardium of mice treated by Folium Isatidis, which characteristic differed from the other groups.3 Some results of viral isolation were positive on day 3 post-infection in infected control group and three treated groups. The positive ratio of viral isolation was increasing increased from day 5 to day 10 post-infection in four group, and then was gradually descended since day 14 post-infection. There were no statistical difference among four groups(p>0.05).4 Perform expression levels in infiltrating cells in murine myocardium were determined by SABC immunohistochemistry. Perforin-positive cells were demonstrated in murine hearts from day 5 to 21 post-infection by immunohistochemistry.
