Objective To study the efficiency of the gene transfer of the gluconoylated polylysine mediating the pCMV 13 Gal(report gene)gene transfer to the A549 cell line and to observe the effect of chloroquine on the efficiency of the gene transfer of the gluconoylated polylysine and to compare the efficiency of gene transfer of the gluconoylated polylysine with that of the Escort liPosome. Escort-transferrin complexes. Methods Applied the Escort 1iposome Escort-transferrin the synthetic gluconoylated polylysine (in the presence of Various concentration of the chloroquine) to mediate the pCMVβ-Gal gene transfer to the A549 cells and analysed quantitatively the expression activity of the f3 galactosidase. Results The gluconoylated polylysine can't mediate the pCMVβ-Gal gene transfer to the A549 cells successfully; The chloroquine can enhance the efficiency of the gene transfer of the gluconoylated polylysine lasmid complex in the A549 cells and when the concentrations of chloroquine was 100 uM, the highest efficiency of gene transfer can be gotten; The Esocrt liposome Escort transferrin complexes can mediate the pCMVβ-Gal gene transfer to the A549 cells successfully. Conclusions The gluconoylated polylysine can't meditat the pCMVβ-Gal gene transfer to the A549 cells, but when the noted concentrations(1O0itM) of chloroquine added, it can mediate the pCMVβ-Gal gene transfer to the A549 cells. The Escort 1iposome, Escort-transferrin complexescan mediate the pCMVβ-Gal gene transfer to the A549 cells successfully.
