The experimental study of high fat MKR mice model:Selecting 20pcs MKR mice of 7 weeks old, half one is male and another half is female. They were randomly divided into two group according to blood sugar's level and body wight and also feed on basal diet and high fat diet respectively. At the same time, there are another 20 pcs wild C57BL/6 mice (C57 mice) used for the parallel experiment. And continuing to feed them 4 weeks. After 11 weeks later, To Measuring the blood sugar level in the empty belly situation, glucose tolerance,CRP, TNF-a, IL-6 and so on.2. The experimental study of zuogui compound having the effect on MKR mouse's blood sugar level in the empty belly situation, insulin level and glucose tolerance:30 pcs, seven weeks old MKR rat are randomly divided into MKR mice model group, Chinese medicine treatment group and west medicine positive control group by the level of blood sugar and body weight. And also the blank control groups of C57 are setted up, each group have 10 pcs rats, using high fat diet to feed them for 4 weeks and till week 11 all of each group will start to gavage. The Chinese medicine group are given the zuogui compound decoction for gavage treatment for 30days and west medicine positive control group are give Rosiglitazone Maleate Tablets for the same treatment. At the same time, MKR mice model and C57 group are given distilled water for gavage treatment for 30 days, once daily. Speciments are collected after 30 days and To Measuring the blood sugar level in the empty belly situation, glucose tolerance,CRP, TNF-a, IL-6 and so on. To test the expression of the proteins of nuclear factorκB (NF-κB) and its upstream activator IKB kinase (3 (IKKβ) protein, p38MAPK and c-Jun N-terminal kinase (JNK) of the organization of Insulin cells (Skeletal muscle, fatand, liver). All the data are analyzed statistically with SPSS 16.0 software.Results:1. MKR mice of high fat group have the obivious pathological changes in the asipose tissue, skeletal muscle and liver after 11 weeks. The level of blood sugar and serum isulin, the amount of CRP, TNF-a, IL-6 increase significantly comparing to the wide C57 mice and also the glucose tolerance is abnormal. Conparing to the MKR mice of basal diet group, which have the deep increasing and the difference have the statistical significant. (P<0.01).2. MKR mice of Zuogui compound group, Has significantly reduced the level of blood sugar and blood insulin, the amount of serum CRP, TNF-a, IL-6 levels, Control the expression level of proteins of NF-κB and its upstream activator IKKβprotein, p38MAPK and JNK of Skeletal muscle, adipose tissue, liver.The difference have the statistical significant comparing to the high fat model. (P<0.01 or P<0.05).Conclusions:1. The control experiment of MKR mice and C57 mice show that MKR mice have the obvious phenomenon of blood sugar level in the empty belly situation and serum insulin increasing, the glucose tolerance is abnormal.while the high-fat feeding can further aggravate insulin resistance.2. Zuogui compound has some functions of lowing blood sugar level, improving glucose tolerance and hyperinsulinemia, reducing inflammatory factors and improving the insulin target organs damage of the adipose tissue, skeletal muscle, liver and so on.3. Zuo gui compound Improve type 2 diabetes of mechanisms maybe to Monitoring NF-κB signaling pathway to Containing Signal elements Exception Express of Upstream and downstream, Relieve the low status of the organization of the inflammation. In order to improve the insulin resistance. Prevent diabetic complications.
