Objective: To research the chemical constituents of the leaves of Capparis spinosal L..To extract and separate the alkaloids from the leaves of Capparis spinosal L..To determinate the contents of stachydrine in different parts of the Capparis spinosa L.. Methods: The chemical constituents of the leaves of Capparis spinosal L. were preliminarily studied by using the methods of systematic pretest and individual pretest. The alkaloids from the leaves of Capparis spinosal L. were extracted, separated and identified by means of TLC, CC and spectral analysis. The contents of stachydrine in different parts of the Capparis spinosa L. were determinated by HPLC. Results: The leaves of Capparis spinosal L. contain alkaloids, flavone, carbohydrates, amino acid, saponins, essential oils, phenols, tannins, organic acids, and steroids. And 3 alkaloids were afforded. Their structures were elucidated by means of Nuclear Magnetic Resonance Spectroscopy(NMR). They are Stachydrine(compound-1), Capparispine(compound-2) and Cadabicine26-O-β-D-glucosidehydrochloride(compound-3). Stachydrine reference substance of the standard curve for Y=1.3311X-0.1128. A good linear ranging of the stachydrine was 2.99~20.94μg. Within the linear range showed good linear relation(r=0.9997), and the average recoveries were 96.1%(RSD=1.53%). Conclusion: The chemical constituents of the leaves of Capparis spinosal L. were preliminarily studied, which will be helpful to the pharmacognosy identification of Capparis spinosal L.. A rout of getting stachydrine is afforded, and the contents of stachydrine in different parts of the Capparis spinosa L. were determinated, which will be valuable to the quality control and pharmacology research.
