Objective: 1. To study on the extraction processes of polysaccharide from Brassica rapa L. and determine the content. 2. To study on the antioxidation activities of Brassica rapa L. polysaccharides samples in vitro. 3. To study on restraining the cancer cell of the Lewis mouse of the polysaccharide from Brassica rapa L. Methods: 1. Use the L9(34) orthogonal design to arrange experiment at the base of the single factor experiment's foundation and determine the polysaccharide's content by the anthrone-sulfuric acid method. 2. The capability of the polysaccharides to deoxidization was determined through the methods of Prussian Blue, the capability of the polysaccharides to eliminate antioxidation activities was determined through eliminating DPPH·, the capability of the polysaccharides to eliminate·OH was determined through sodium salicylate method, the capability of the polysaccharides to eliminate O2- was determined through pyrogallol auto oxidation. 3. The anti-tumor effects of polysaccharide from Brassica rapa L .were testd in mice with transplanted tumorsarcom Lewis, the tumor inhibition rate, life span. speen index and the lymphocyte transformation function in vitro are detemined. Results: 1. By L9(34) orthogonal design and analyses of variance , the extraction processes of polysaccharides from Brassica rapa L.: the extraction temperature is 90℃, the extraction time is 2 h, the ration of solidmaterial to liquid is1:30(g/ml), the extraction times is 3, the content reached 10.94%. 2. The antioxidation of Brassica rapa L.polysaccharide: BSP, had the ability of reducing ability and scavenging oxygen free radicals, hydroxyl free radical, DPPH·radical. But DPPH·radical scavenging ability and reducing ability of BSP-1 were higher than BSP , oxygen free radicals scavenging ability and hydroxyl free radical of BSP were higher than BSP-1. 3. The tumor weight of Brassica rapa L. polysaccharides groups has statistical significance compared to NS group(P<0.01), the spleen index of polysaccharides groups has statistical significance to NS group (P<0.05), survival days of polysaccharides groups has statistical significance compared to NS group (P < 0.01). This drug can also enhance the lymphocyte transformation function in vitro. Conclusion: 1. The method is simple, stability,repeatability and precision are comply with the requirements, and has a wide range of applicabilty. 2. The results of the antioxidation activities showed that BSP,BSP-1 had different antioxidant activities: deoxidization , had better effects of elimination of O2-,·OH,DPPH. 3. Brassica rapa L. polysaccharides can increase the tumor inhibitory rate,the spleen index,life prolonge rate. The drug can increase the lymphocyte transformation in vitro. |
