Brassica rapa L.,planted in the Qingzang-Tibet Plateau,tasted as pungent and mild in character.For thousands of years,it has become a medicinal,edible and forage plant in the Qinghai-Tibet Plateau,for alleviate the altitude sickness of local people and livestock.Studies have shown that polysaccharides,flavonoids,polyphenols,triterpenoids,saponins and other substances contained in Brassica rapa L.are its main functional components,and these compounds show functional activities such as hypoxia tolerance,fatigue resistance,radiation resistance and immune regulation.The characteristic components,such as glucosinolates and isothiocyanates,have remarkable anti-tumor,anti-virus and antibacterial activities.It is found that the crude extract of the root of Brassica rapa L.has certain effects of relieving acute exercise fatigue and brain protection,and can effectively alleviate the peripheral or central nervous system damage caused by oxidative stress and inflammation.However,there is no report on the effect of Brassica rapa L.in intervening chronic fatigue.As a negative state,fatigue can affect anyone.Chronic fatigue will be formed with the extension of fatigue symptoms,and it is often accompanied by various diseases.Chronic fatigue is a debilitating syndrome with unknown etiology.At present,the causes of chronic fatigue mainly include the following aspects: abnormal immune system,abnormal central nervous system function,abnormal neuroendocrine system and abnormal muscle energy metabolism.In addition,intestinal flora also plays an important role in chronic fatigue.The imbalance of intestinal flora and the damage of intestinal barrier will induce many diseases including chronic fatigue.Because the pathogenesis of chronic fatigue has multiple targets,intervention with herbs rich in various active compounds is becoming a mainstream means to combat chronic fatigue.Therefore,in this study,mice with fatigue induced by continuous swimming were used to simulate the chronic fatigue state,and the extract of the roots of Brassica rapa L.was used to intervene.The main research contents are as follows:(1)In this experiment,27 kinds of organic acids,15 kinds of glucosinolates,12 kinds of glucosides,16 kinds of flavones,5 kinds of flavones,3 kinds of phenols,6 kinds of esters and7 kinds of isothiocyanates were identified by fractional extraction,which are homogeneous with the standard samples and have reference significance.The order of antioxidant capacity in vitro is as follows: AEB > EAET > SBET > PET > CET > CHL.Molecular docking shows that sulforaphane has good binding ability with Nrf2,trans-6-shogaol and MAPK,triandrin and PGC-1α.It is speculated that AEB can improve antioxidant capacity,stimulate immune activity and promote mitochondrial function.(2)Exploring the intervention effect of AEB on peripheral fatigue of chronic fatigue mice,it is found that AEB can significantly improve the exercise state of fatigue mice,regulate the fatigue-related metabolites such as BUN and LD in vivo,and regulate the activities of enzymes such as LDH and CK.It also regulates NAD+/NADH to promote ATP synthesis and maintain the level of glycogen in liver muscle.And by regulating the balance between mTOR and AMPK,it can promote cell energy metabolism,and promote the expression of PGC-1α/TFAM in the downstream pathway,and improve mitochondrial function and biogenesis.By regulating Nrf2/HO-1 pathway,the synthesis of antioxidant enzymes SOD,CAT and GPx in vivo was promoted,the antioxidant defense system was strengthened,and the oxidative stress was reduced.And effectively promote the growth of skeletal muscle fibers,reduce injury and effectively reduce leukocyte infiltration.(3)Exploring the intervention effect of AEB on central fatigue of chronic fatigue mice,it was found that the performances in Y-maze test have been enhanced significantly.Then,AEB increased the neurotransmitter DA in hypothalamus and striatum,inhibited 5-HT in hypothalamus and neurotransmitters Glu and GABA in hypothalamus and striatum,and optimized the mental state of chronic fatigue mice.AEB up-regulated the expression of SIRT1/PGC-1α/TFAM pathway in hippocampus of fatigue mice,and maintained the mitochondrial function of hippocampus in mice.By establishing the model of HT22 cells induced by Glu,it is found that AEB can effectively reduce the accumulation of ROS induced by Glu,maintain the stable growth of cells,effectively maintain the mitochondrial membrane potential and mitochondrial morphology stability of cells,and promote mitochondrial biogenesis by increasing AMPK/PPARγ expression.It shows that mitochondria may be one of the potential targets for AEB to intervene chronic fatigue.(4)Exploring the effect of AEB on cytokines in mice with chronic fatigue,it was found that by inhibiting the TLR4/NF-κB signaling pathway in various tissues and organs,the roots of Aralia elata regulated the expression of proinflammatory cytokines such as IL-6,TNF-α,IL-1β,IL-2 and IL-10,and alleviated the inflammation of muscles,striatum and colon.And by regulating TFG-β,VEGF and PDGF-BB,it may have a protective effect on muscle fiber regeneration and barrier function of the body.(5)The effect of the root of Daphne giraldii on the intestinal flora of mice.The roots of Daphne giraldii increased the abundance of beneficial bacteria such as Firmicutes,Roseburia,Faecalibacterium,Clostridioides,Ruminococcus,Lactobacillus and Bacteroides in the intestine of chronic fatigue mice.The abundance of harmful bacteria such as Proteobacteria,Enterococcus,Mucispirillum,Pseudomonas and Enterobacteriaceae was inhibited.Promote the production of acetic acid,propionic acid,isovaleric acid and total short-chain fatty acids.By up-regulating the expression of Occludin and HO-1,the colon barrier was protected,the level of serum endotoxin was reduced,and PGC-1α/TFAM was regulated.Enriching the function of the flora,it is predicted that the root of Daphne odora can improve the vitamin B12 synthesis pathway in mice.(6)Anti-fatigue effect of compound vitamin beverage of Brassica rapa L.The optimal taste formula of the beverage was determined by single factor and response surface optimization experiments,namely,80% of the original juice,12% of white sugar and 0.50% of citric acid,and vitamin B3,vitamin B6,vitamin B12,18mg/L,1.6 mg/L and 1.6 μg/L were compounded respectively.It can effectively improve the exercise ability of fatigue mice,optimize fatigue metabolites and improve glycogen storage.
