The Multi-drug resistance of Candida albicans is the main reason for the failure clinical therapy. The subject of using gene knockout technology and systematic study of the MIR1 features:The Ura-Blaster strategy will MIR1 two alleles from the genome of C. albicans knockout, gene deletion strains constructed MIR1 then used to study the MIR1 functions. The results showed:MIR1 gene knockout slow down the growth of Candida albicans; by biofilm growth kinetics determination (XTT method), laser scanning confocal microscopy (CSLM) observation of biofilms and cell surface hydrophobicity (CSH) and other measured method was investigated MIR1 gene decreased biofilm formation and the ability of hyphae. MIR1 gene in Candida albicans biofilm formation plays an important role. Investigation MIR 1 gene on drug sensitivity:The results show that MIR1 gene fluconazole, ketoconazole and terbinafine drug sensitivity decrease, Spot assay and Filter disk assay results. Rhodamine 6G results showed:MIR1 knockout of Candida albicans drug efflux capacity increases, the drug efflux pump enhancements; but the detection of a resistance gene was found that transcription did not change significantly. Examined the level of intracellular ATP was found:MIR1 gene knockout, elevated levels of intracellular ATP; study found the level of mitochondrial membrane potential:MIR1 gene knockout, a direct indicator of mitochondrial function-increased mitochondrial membrane potential. Endogenous reactive oxygen species (ROS) exceeds a certain concentration will itself produce oxidative damage, is the major inhibitory azole drugs, one of the factors; the same time the mitochondrial respiratory chain is the endogenous major source of reactive oxygen species was found:MIR1 knock In addition, the endogenous activity of Candida albicans to increase oxygen levels lower than the parent strain. In summary, MIR1 gene knockout of Candida albicans causes reduced susceptibility to drugs, partly because drug efflux pump energy increases, so that enhancements; the other reason is that knocking endogenous sterilization lower than the increase of reactive oxygen species parental strains. The topic from various aspects, levels stated, MIR1 impact of Candida albicans induced drug resistance.
