| The Multi-drug resistance of Candida albicans is the main reason for the failure clinical therapy .Now the sequencing results of the Candida albicans genome has presented and could be gained in public database.This study find that a novel gene IPF18207 in Candida albicans shows highly homology with gene DAP1 in Saccharomyces cerevisiae after blastn and blastp.It has proved that in Saccharomyces cerevisiae Dap1p could influence the stability of Erg11p,a key enzyme in ergosterol biosynthetic pathway and a target enzyme of many azoles drugs. So Dap1p could indirectly influence the sensitivity to azoles drugs in Saccharomyces cerevisiae.Ipf18207p may have the similar function in Candida albicans. IPF18207 gene knock-out strain was constructed with Ura-Blaster strategies,and verificated by PCR and southern blotting.This strain was used to analyze the functiong of IPF18207 gene.The results revealed : IPF18207 gene did not influenced growth curve and drug (Amphotericin B, itraconazole, fluconazole, ketoconazole, Itraconazole and Terbinafine )sensitivity significantly,but increased the sensitivity of methyl methanesulfonate (MMS). IPF18207 gene knock-out strain could tolerate higher concentration and generate more ROS than wild strain.The results indicats that product encoded by IPF18207 gene may be involved in oxidative stress. |
