The definition of meridian doctrine can be described as TCM selective effect or particular effect to the particular part of human body.Tianma which is the dried rhizome of Gastrodia elata BL. is an important and notable Chinese herbal medicine. It has been used for over one thousand years in China. Tianma is the most important drug for the treatment of Hyperactivity of liver-YANG syndrome (HLYS), liver within the dynamic wind.In the first, this study has established a method for the determination of gastrodin which is the main effect components of Tianma in SHR rat tissues. Through the method, the meridian doctrine study on gastrodin after oral administration of Gastrodia and extract composed of Gastrodia and Uncaria Rhynchophylla (Miq.) Jacks in SHR rat tissues. By discussing the role of compound compatibility of the process of Gastrodia effects in rats, this study suggested the Preliminary meridian doctrine theory of Gastrodia and investigated the regularity of compatibility of drug couple in the meridian doctrine.1. The purpose of this study was to establish a method for the determination of gastrodin which is the effective constituents of Tianma in SHR rat tissues.The basic contents of this study are as follows:SHR rats were used the method of oral administration with normal saline, the rat liver and kidney were removed after administrated of normal saline. After drawn and treated by adding precipitation reagent and the supernatant, the tissue samples were dried by N2.The residues which were extracted in solvent were detected by High Performance Liquid Chromatography (HPLC) directly.RESULTS:The rat liver and kidney were removed after oral administration. The samples were added normal saline to obtain the homogenate solution. The tissue samples which were drawn and treated by adding methanol and the supernatant were dried by N2. After extracting in water,the residues were detected by HPLC directly. The analysis was carried on an analytical column Phenomenence Luna C18 (5μm,250×4.6 mm). The mobile phase consisted of acetonitrile-1%H3PO4 (3:97) and the flow rate was 1.0 mL·min-1 The UV detection wave length was at 220 nm and the temperature of column was kept at 30℃.CONCLUSION:The method is simple, stable and easy for operating. It is shown to be suitable for determination of gastrodin in SHR rat tissues.2. The purpose of this study was to study Meridian doctrine on determination of Gastrodin in tissues of SHR rats and its change before and after compatibility of Gastrodia with Uncaria Rhynchophylla.SHR rats were used the method of oral administration with Tianma suspension and Tianmagouteng Decotion (TD). Trough determined the pretreatment method, the contents of gastrodin were detected and analyzed in the those tissues of liver and kidney after oral administration.RESULTS:The SHR rats were used the method of oral administration with Tianma suspension and Tianmagouteng Decotion (TD) and were detected the content of gastrodin in the tissues. The concentration of gastrodin in the kidney was higher than the liver. Both before and after compatibility of Gastrodia with Uncaria, gastrodin distribution in the kidney was higher than the liver. The concentration of Gastrodin reached peaks of distribution twice in the liver, while in the kidney did not appear this situation. In the liver tissues, after compatibility of Tianma with Uncaria, its concentration in gastrodin maximum points (60min) increased in the concentration distribution, while the double-peak phenomenon disappeared. In the kidney tissues, the distribution did not show changes significantly as well as liver tissues, but at the concentration maximum time points (60min) the concentration of gastrodin was lower than before. After compatibility of Gastrodia with Uncaria, the concentration increased in the liver tissues and decreased in the kidney tissues. The data suggested that Uncaria could enhance gastrodin to pass through liver channel.CONCLUSION:After compatibility of Gastrodia with Uncaria, the concentration of gastrodin changed significantly in the liver and kidney tissues. It accelerated the absorption distribution of gastrodin and rate of elimination. The results showed that Uncaria could influence notably the distribution of gastrodin which is the main effect components of Gastrodia, accelerated the distribution in the liver tissues and increased the speed of elimination. The results suggested that Uncaria could enhance gastrodin to pass through liver channel and play the role of accelerating the distribution and metabolism of gastrodin reducing length of staying time in the tissues. |
